The disclosure provides novel DNA sequences, synthesized in the 3′ to 5′ direction using topoisomerase, comprising a sequence of oligomeric cassettes, wherein each oligomeric cassette comprises a topoisomerase recognition sequence and an informational sequence, and methods for making and using the same.

32 . A DNA sequence comprising an informational series of oligomeric cassettes, wherein each oligomeric cassette comprises a topoisomerase recognition sequence and an informational sequence.
33 . The DNA sequence of claim 32 wherein the topoisomerase recognition sequence is recognized by Vaccinia topoisomerase I or by SVF topoisomerase 1.
34 . The DNA sequence of claim 32 wherein the topoisomerase recognition sequence is 5′-(C/T)CCTT-3′ or 5′-CCCTG-3′.
35 . The DNA sequence of claim 34 wherein the topoisomerase recognition site is 5′-CCCTT-3′.
36 . The DNA sequence of claim 32 wherein the informational sequence of each cassette is selected from at least two different sequences.
37 . The DNA sequence of claim 36 wherein the informational sequence of each cassette is a sequence of 3 to 12 nucleotides.
38 . The DNA sequence of claim 36 wherein the informational sequence of each cassette is selected from two different sequences, one of which corresponds to a “1” and one of which corresponds to a “0”, so that the informational series of oligomeric cassettes contains information in binary code form.
