Abstract

The present invention relates to polymorphs of the hydrochloride salt of 5-{2-[({8-[(2,6-dimethylbenzyl)amino]-2,3-dimethylimidazo[1,2-a]pyridine-6-yl}carbonyl)-amino]ethoxy}-5-oxopentanoic acid (linaprazan glurate), more specifically Form 1 and Form 2 of the HCl salt of linaprazan glurate. The invention also relates to a process for the preparation of such polymorphs, to pharmaceutical compositions comprising such polymorphs, and to the use of these polymorphs in the treatment or prevention of gastrointestinal inflammatory diseases or gastric acid related diseases, in particular erosive gastroesophageal reflux disease (eGERD).

Claims (15)

1. A method for treating a gastrointestinal inflammatory disease or a gastric acid related disease in a subject in need thereof, the method comprising administering to the subject a therapeutically effective amount of a crystalline HCl salt of linaprazan glurate, wherein the gastrointestinal inflammatory disease or the gastric acid related disease is gastritis, gastroesophageal reflux disease (GERD), erosive gastroesophageal reflux disease (eGERD), H. pylori infection, Zollinger-Ellison syndrome, peptic ulcer disease (including gastric ulcers and duodenal ulcers), bleeding gastric ulcer, symptoms of gastroesophageal reflux disease (including heartburn, regurgitation and nausea), gastrinoma or acute upper gastrointestinal bleeding, wherein the crystalline HCl salt of linaprazan glurate is an anhydrate which is Form 1, having an XRPD pattern, obtained with CuKα1-radiation, with at least two peaks at °2θ values selected from the list consisting of 3.8±0.2, 9.1±0.2, 13.8±0.2, 14.0=0.2, 20.0±0.2, 22.9±0.2, 23.4±0.2, 24.4±0.2, 24.6±0.2 and 26.7±0.2.

2. The method of claim 1 , wherein Form 1 has an XRPD pattern, obtained with CuKα-radiation, with at least peaks at °2θ values of 20.0=0.2, 24.4±0.2, 24.6=0.2 and 26.7=0.2.

3. The method of claim 1 , wherein Form 1 has an XRPD pattern, obtained with CuKα-radiation, with at least peaks at °2θ values of 9.1=0.2, 13.8=0.2, 20.0±0.2, 23.4=0.2, 24.4=0.2, 24.6±0.2 and 26.7±0.2.

4. The method of claim 1 , wherein the crystalline HCl salt of linaprazan glurate is Form 1, having an XRPD pattern, obtained with CuKα-radiation, substantially as shown in FIG. 1 .

5. The method of claim 1 , wherein Form 1 has a DSC curve comprising an endotherm between about 230° C. and about 240° C., such as at approximately 233° C.

A method for treating a gastrointestinal inflammatory disease or a gastric acid related disease in a subject in need thereof, the method comprising administering to the subject a therapeutically effective amount of a crystalline HCl salt of linaprazan glurate, wherein the gastrointestinal inflammatory disease or the gastric acid related disease is gastritis, gastroesophageal reflux disease (GERD), erosive gastroesophageal reflux disease (eGERD), H. pylori infection, Zollinger-Ellison syndrome, peptic ulcer disease (including gastric ulcers and duodenal ulcers), bleeding gastric ulcer, symptoms of gastroesophageal reflux disease (including heartburn, regurgitation and nausea), gastrinoma or acute upper gastrointestinal bleeding, wherein the crystalline HCl salt of linaprazan glurate is a non-stoichiometric hydrate which is Form 2, having an XRPD pattern, obtained with CuKα1-radiation, with at least two peaks at °2θ values selected from the list consisting of 7.1±0.2, 9.9=0.2, 10.2±0.2, 15.0=0.2, 15.7=0.2, 22.6±0.2, 22.8=0.2 and 25.0±0.2.

7. The method of claim 6 , wherein Form 2 has an XRPD pattern, obtained with CuKα1-radiation, with at least peaks at °2θ values of 7.1±0.2, 15.0±0.2, 22.6=0.2 and 25.0=0.2.

8. The method of claim 6 , wherein Form 2 has an XRPD pattern, obtained with CuKα1-radiation, with at least peaks at °2θ values of 7.1±0.2, 9.9±0.2, 10.2±0.2, 15.0±0.2, 15.7±0.2, 22.6±0.2, 22.8±0.2 and 25.0±0.2.

9. The method of claim 6 , wherein the crystalline HCl salt of linaprazan glurate is Form 2, having an XRPD pattern, obtained with CuKα-radiation, substantially as shown in FIG. 2 or FIG. 3 .

10. The method of claim 6 , wherein Form 2 has a DSC curve comprising an endotherm between about 175° C. and about 185° C., such as at approximately 180° C.

11. The method of claim 1 or claim 6 , wherein the crystalline HCl salt of linaprazan glurate has a crystallinity of greater than 99%.

12. The method of claim 1 , wherein the gastrointestinal inflammatory disease or the gastric acid related disease is erosive gastroesophageal reflux disease (eGERD).

13. The method of claim 1 , wherein the gastrointestinal inflammatory disease or the gastric acid related disease is H. pylori infection.

14. The method of claim 1 , wherein the gastrointestinal inflammatory disease or the gastric acid related disease is gastroesophageal reflux disease (GERD).

15. The method of claim 6 , wherein the gastrointestinal inflammatory disease or the gastric acid related disease is gastroesophageal reflux disease (GERD), erosive gastroesophageal reflux disease (eGERD) or H. pylori infection.
