Abstract

A method for producing a three-dimensional tissue construct comprising mature adipocytes, comprising incubating cells comprising at least fat-derived stein cells in the presence of one or more fatty acids selected from the group consisting of erucic acid, elaidic acid, oleic acid, palmitoleic acid, myristoleic acid, phytanic acid, and pristanic acid.

Claims (18)

1 . A method for producing a three-dimensional tissue construct comprising mature adipocytes, comprising incubating cells comprising at least fat-derived stem cells in the presence of one or more fatty acids selected from the group consisting of erucic acid, elaidic acid, oleic acid, palmitoleic acid, myristoleic acid, phytanic acid, and pristanic acid.

2 . The method according to claim 1 , comprising incubating the cells in the presence of a TGFβ type I receptor inhibitor.

The method according to claim 2 , wherein the incubating in the presence of a TGFβ type I receptor inhibitor is incubating in a culture medium comprising the TGFβ type I receptor inhibitor, and a content of the TGFβ type I receptor inhibitor in the culture medium is 1 μM or more and 10 μM or less.

4 . The method according to claim 1 , wherein the cells comprising at least fat-derived stem cells include no mature adipocyte.

5 . The method according to claim 1 , wherein the fat-derived stem cells are bovine-derived.

6 . The method according to claim 1 , wherein the incubating is performed for 96 hours or more and 384 hours or less.

7 . The method according to claim 1 , comprising incubating the cells together with a fragmented extracellular matrix component in the presence of a TGFβ type I receptor inhibitor.

8 . The method according to claim 7 , wherein the fragmented extracellular matrix component is disposed in a gap among the cells in the three-dimensional tissue construct.

9 . The method according to claim 7 , wherein the fragmented extracellular matrix component is a fragmented collagen component.

10 . The method according to claim 7 , further comprising a step of bringing the cells into contact with the fragmented extracellular matrix component in an aqueous medium before incubation.

A method for promoting differentiation of fat-derived stem cells, comprising incubating cells comprising at least fat-derived stem cells in the presence of one or more fatty acids selected from the group consisting of erucic acid, elaidic acid, oleic acid, palmitoleic acid, myristoleic acid, phytanic acid, and, pristanic acid.

12 . The method according to claim 11 , comprising incubating the cells in the presence of a TGFβ type I receptor inhibitor.

The method according to claim 12 , wherein the incubating in the presence of a TGFβ type I receptor inhibitor is incubating in a culture medium comprising the TGFβ type I receptor inhibitor, and a content of the TGFβ type I receptor inhibitor in the culture medium is 1 μM or more and 10 μM or less.

14 . The method according to claim 11 , wherein the fat-derived stem cells are bovine-derived.

15 . The method according to claim 11 , wherein the incubating is performed for 96 hours or more and 384 hours or less.

16 . The method according to claim 11 , comprising incubating cells comprising at least fat-derived stem cells together with a fragmented extracellular matrix component in the presence of a TGFβ type I receptor inhibitor.

17 . The method according to claim 16 , wherein the fragmented extracellular matrix component is a fragmented collagen component.

18 . The method according to claim 16 , further comprising a step of bringing the cells into contact with the fragmented extracellular matrix component in an aqueous medium before incubation
