Abstract

The invention provides a process of purification of antibody or fusion protein from protein mixture comprising product and process related impurities. The process provides the use of hydroxyapatite chromatography for the separation of low molecular weight impurities and basic variants. In addition, invention further provides a scalable purification process to remove product and process related impurities.

Claims (41)

1. A process of purifying a protein of interest from a protein mixture comprising the protein of interest and a low molecular weight (LMW) variant 2H1L or HH, the process comprising:

a) loading the protein mixture onto Ceramic Hydroxy apatite (CHT) column with a loading buffer;

b) washing the CHT column with a washing buffer; and

c) eluting by a linear gradient with an eluting buffer, thereby producing an eluted protein mixture comprising a purified protein of interest;

wherein the washing buffer is sodium phosphate buffer and the eluting buffer is sodium phosphate buffer;

wherein the eluting buffer comprises sodium phosphate concentration from about 24 mM to about 104 mM; and

wherein the purified protein of interest is substantially pure and 2H1L is less than 3% measured by CE-SDS or HH is less than 0.2% measured by CE-SDS and wherein the purified protein of interest is substantially pure.

2. The process of claim 1 , the eluted protein mixture eluted comprises low 2H1L in an amount less than 2.6% or low HH in an amount less than 0.2%.

3. The process of claim 1 , wherein the concentration of phosphate is increased gradually from about 40 mM to 96 mM.

4. The process of claim 1 , wherein eluting by a linear gradient with an eluting buffer is performed for 19 CV.

5. The process of claim 1 , wherein the eluted protein mixture in step (c) is collected from starting at an ascending value of absorbance UV280 at about 1.5 AU/cm and ending at a descending value of about 1.5 AU/cm.

6. The process of claim 1 , wherein the eluted protein mixture in step (c) is collected from starting at an ascending value of absorbance UV280 at about 1.0 AU/cm and ending at a descending value of about 1.5 AU/cm.

7. The process of claim 1 , wherein the loading buffer has same conductivity and pH as the washing buffer.

8. The process of claim 1 , wherein each of the loading buffer, the washing buffer, and the eluting buffer has pH 7.0±0.2.

9. The process of claim 1 , wherein each of the loading buffer, the washing buffer, and the eluting buffer does not have any additives selected from sodium chloride, calcium chloride, and glycine.

10. The process of claim 1 , wherein the protein of interest is an IgG1 antibody that binds to IgE.

11. The process of claim 10 , wherein the IgG1 antibody is omalizumab.

12. The process of claim 1 , wherein purity of the purified protein of interest is more than 90% as measured by SE-HPLC.

13. The process of claim 1 , wherein the eluted protein mixture eluted comprises about 1.5% or less of 2H1L or about 0.06% or less of HH.

14. The process of claim 1 , wherein purity of the purified protein of interest is more than 70% as measured by CEX-HPLC.

15. A process of purifying a protein of interest from a protein mixture comprising a protein of interest and one or more low molecular weight (LMW) variants, the process comprising:

a) loading the protein mixture onto Ceramic Hydroxy apatite (CHT) column;

b) washing the CHT column with a washing buffer;

c) eluting the purified protein of interest by linear gradient with an eluting buffer, thereby producing a purified protein of interest;

wherein the washing buffer is sodium phosphate buffer and the eluting buffer is sodium phosphate buffer at a concentration of about 24 mM to about 104 mM;

wherein the purified protein of interest is substantially pure; and

wherein the one or more low molecular weight variants are present in an amount below 0.4%, as analysed by SE-HPLC Analysis.

16. The process of claim 15 , wherein the purified protein of interest comprises a reduced amount of the one or more LMW variants by at least 95% as measured by SE-HPLC analysis.

17. The process of claim 15 , wherein the one or more LMW variants is selected from LC, HC, HH, and 2H1L.

18. The process of claim 17 , wherein the HC is present in an amount below 0.4% measured by CE-SDS 0.4%.

19. The process of claim 15 , wherein the LC is present in an amount below 0.4% measured by CE-SDS.

20. The process of claim 15 , wherein the concentration of phosphate in the eluting buffer is increased gradually from about 40 mM to 96 mM.

21. The process of claim 15 , wherein the linear gradient with the eluting buffer comprises a phosphate concentration from about 10% to 24%.

22. The process of claim 15 , wherein eluting the purified protein of interest by linear gradient with an eluting buffer is performed for 19 CV.

23. The process of claim 15 , wherein the eluted protein mixture in step (c) is collected from starting at an ascending value of absorbance UV280 at about 1.5 AU/cm and ending at a descending value of absorbance UV280 at about 1.5 AU/cm during the elution.

24. The process of claim 15 , wherein the eluted protein mixture in step (c) is collected from starting at an ascending value of about 1 . 0 AU/cm and ending at a descending value of absorbance UV280 about 1.5 AU/cm during the elution.

25. The process of claim 15 , wherein the loading buffer and has same conductivity and pH as the washing buffer.

26. The process of claim 15 , wherein each of the loading buffer, the washing buffer, and the eluting buffer has pH 7.0±0.2.

27. The process of claim 15 , wherein each of the loading buffer, the washing buffer, and the eluting buffer does not have any additives selected from sodium chloride, calcium chloride, and glycine.

28. The process of claim 15 , wherein the protein of interest is an IgG1 antibody that binds to IgE.

29. The process of claim 28 , wherein the IgG1 antibody is omalizumab.
