Abstract

Provided herein are compositions and methods for producing milk proteins, which allow for safe, sustainable and humane production of milk proteins for commercial use, such as use in food compositions. The disclosure provides recombinant fusion proteins comprising at least first protein and a second protein, wherein at least one of the first protein and the second protein is a milk protein, or fragment thereof. The disclosure also provides methods for producing the recombinant fusions proteins, and food compositions comprising the same.

Claims (29)

1. A plant cell comprising:

a) a heterologous casein protein; and

b) a heterologous Fam20C kinase protein;

wherein the heterologous casein protein is intracellularly phosphorylated by the Fam20C kinase protein: wherein the plant cell accumulates higher quantities of the heterologous casein protein than a control plant cell lacking the heterologous Fam20C kinase protein.

2. The plant cell of claim 1 , wherein the heterologous casein protein is phosphorylated at a Ser-X-Glu/pSer motif by the heterologous Fam20C kinase protein.

3. The plant cell of claim 1 , wherein the plant cell is from a dicot.

The plant cell of claim 3 , wherein the dicot is selected from the group consisting of arabidopsis , tobacco, tomato, potato, sweet potato, cassava, alfalfa, lima bean, pea, chickpea, soybean, carrot, strawberry, lettuce, oak, maple, walnut, rose, mint, squash, daisy, quinoa , buckwheat, mung bean, cow pea, lentil, lupin, peanut, fava bean, French bean, mustard, and cactus.

5. The plant cell of claim 3 , wherein the dicot is soybean.

6. The plant cell of claim 1 , wherein the plant cell is from a monocot.

7. The plant cell of claim 6 , wherein the monocot is selected from the group consisting of maize, rice, oat, wheat, barley, and sorghum.

8. The plant cell of claim 1 , wherein the heterologous Fam20C protein comprises SEQ ID NO: 821, or a sequence at least 90% identical thereto.

9. The plant cell of claim 1 , wherein the heterologous Fam20C protein comprises SEQ ID NO: 821.

10. The plant cell of claim 1 , wherein the heterologous Fam20C protein is encoded by a nucleic acid sequence comprising SEQ ID NO: 820, or a sequence at least 90% identical thereto.

11. The plant cell of claim 1 , wherein the heterologous Fam20C protein comprises amino acids 94-586 of SEQ ID NO: 821, or a sequence at least 90% identical thereto.

12. The plant cell of claim 1 , wherein the heterologous Fam20C protein comprises amino acids 94-586 of SEQ ID NO: 821.

13. The plant cell of claim 1 , wherein the heterologous casein protein is expressed from a nucleic acid encoding the heterologous casein protein, and operably linked to a seed-specific promoter.

14. The plant cell of claim 13 , wherein the seed-specific promoter is selected from the group consisting of PvPhas, BnNap, AtOle1, GmSeed2, GmSeed3, GmSeed5, GmSeed6, GmSeed7, GmSeed8, GmSeed10, GmSeed11, GmSeed12, pBCON, GmCEP1-L, GmTHIC, GmBg7S1, GmGRD, GmOLEA, GmOLER, Gm2S-1, and GmBBld-II.

15. The plant cell of claim 1 , wherein the heterologous casein protein is expressed from a construct that also encodes the heterologous Fam20C protein.

16. The plant cell of claim 1 , wherein the heterologous casein protein is selected from the group consisting of: α-S1 casein, α-S2 casein, β-casein, κ-casein, and para-κ-casein.

17. The plant cell of claim 1 , wherein the heterologous casein protein is a mature truncated casein, lacking a signal peptide.

18. The plant cell of claim 1 , wherein the heterologous casein protein is within a fusion protein comprising a second milk protein or a zein protein.

The plant cell of claim 18 , wherein the second milk protein is selected from the group consisting of an α-S1 casein, an α-S2 casein, a β-casein, a κ-casein, a para-κ-casein, a (β-lactoglobulin, an α-lactalbumin, a lysozyme, a lactoferrin, a lactoperoxidase, a serum albumin, and an immunoglobulin.

20. The plant cell of claim 1 , wherein the Fam20C kinase protein comprises a plant signal peptide.

21. The plant cell of claim 1 , wherein the heterologous casein protein, the heterologous Fam20C kinase protein, or both, comprise an endoplasmic reticulum retention signal.

22. The plant cell of claim 21 , wherein the endoplasmic reticulum retention signal is the “KDEL” signal (SEQ ID NO: 23).

23. The plant cell of claim 20 , wherein the plant signal peptide is selected from the list consisting of GmSCB1, StPat2l, 2Sss, Sig2, Sig12, Sig8, Sig10, Sig11, and Coixss.

24. The plant cell of claim 20 , wherein the plant signal peptide is Sig10.

25. The plant cell of claim 20 , wherein the plant signal peptide comprises the amino acid sequence of SEQ ID NO: 15.

26. The plant cell of claim 20 , wherein the plant signal peptide is encoded by the nucleic acid sequence of SEQ ID NO: 819.
