DIFFERENTIATION AND ENRICHMENT OF ISLET-LIKE CELLS FROM HUMAN PLURIPOTENT STEM CELLS
Methods for differentiating human pluripotent stem cells into islet-like cells are provided. In certain embodiments, the methods utilize sequential culturing of the human pluripotent stem cells with certain factors to produce islet-like cells. In certain embodiments, the population of cells produced by methods is further enriched for islet-like cells.

17 . A method comprising the steps of:
a) obtaining a plurality of cell clusters, wherein the plurality of clusters comprise: i) clusters having a particle size of 70 microns to 200 microns, ii) clusters having a particle size of less than 70 microns in size, and iii) clusters having a particle size of greater than 200 microns in size; and
b) removing the clusters that have a particle size of less than 70 microns or greater than 200 microns in size from the plurality of cell clusters;
wherein the cell clusters comprise cells that express PDX1 and at least one of insulin, C-peptide, or glucagon.
18 . The method of claim 17 , wherein the cell clusters comprise cells that express insulin.
19 . The method of claim 17 , wherein the cell clusters comprise cells that express glucagon.
20 . The method of claim 17 , wherein said the cells in said cultures are genetically altered.
21 . The method of claim 20 , wherein said genetic alteration enhances therapeutic effect of said cultured cells upon implantation into a subject.
22 . The method of claim 20 , wherein said genetic alteration comprises an effector gene.
