Hydrocortisone acetate suppository formulation for treatment of disease
The present invention relates, in various embodiments, to formulations comprising hydrocortisone and silicon dioxide. In additional embodiments, the invention relates to suppositories comprising hydrocortisone and silicon dioxide. The formulations of the present invention are useful for administration to patients who have gastrointestinal diseases and disorders such as, for example, inflammatory bowel disease.
1. A formulation comprising about 0.5% to about 5% by weight hydrocortisone acetate, about 0.1% to about 5% by weight colloidal silicon dioxide, and a glyceride base comprising lecithin, wherein the formulation has a melting temperature in the range of about 35° C. to about 39° C.
2. The formulation of claim 1 , comprising about 4.5% by weight hydrocortisone acetate and about 0.7% by weight colloidal silicon dioxide.
3. The formulation of claim 1 , wherein the base comprises at least about 85% triglycerides, and further comprises diglycerides and monoglycerides.
4. The formulation of claim 1 , wherein the formulation releases the hydrocortisone acetate upon exposure to rectal fluid.
5. The formulation of claim 1 , wherein the glyceride base further comprises triglyceride fatty acids.
6. The formulation of claim 5 , wherein the fatty acids are saturated C8-C18 fatty acids.
7. The formulation of claim 1 , wherein the glyceride base is a semi-synthetic glyceride base.
8. The formulation of claim 1 , wherein the formulation comprises about 90 mg of hydrocortisone acetate.
9. The formulation of claim 1 , wherein the formulation comprises about 85 to about 95 mg of hydrocortisone acetate.
10. The formulation of claim 1 , wherein the formulation comprises 90 mg of hydrocortisone acetate.
11. The formulation of claim 1 , wherein the formulation comprises 85 to 95 mg of hydrocortisone acetate.
12. The formulation of claim 1 , wherein the formulation comprises about 94.8% w/w of the glyceride base.
13. The formulation of claim 2 , wherein the formulation comprises about 94.8% w/w of the glyceride base.
14. The formulation of claim 13 , wherein the formulation comprises 85 to 95 mg of hydrocortisone acetate.
15. The formulation of claim 14 , wherein the formulation comprises 90 mg of hydrocortisone acetate.
16. The formulation of claim 2 , wherein the formulation is a suppository with a weight of about 2 grams.
17. The formulation of claim 15 , wherein the formulation is a suppository with a weight of about 2 grams.
