Abstract

Embodiments of the invention relate generally to methods to generate microorganisms and/or microorganism cultures that exhibit the ability to produce polyhydroxyalkanoates (PHA) from carbon sources at high efficiencies. In several embodiments, preferential expression of, or preferential growth of microorganisms utilizing certain metabolic pathways, enables the high efficiency PHA production from carbon-containing gases or materials. Several embodiments relate to the microorganism cultures, and/or microorganisms isolated therefrom.

Claims (40)

1. A method for generating a culture of methanotrophic microorganisms exhibiting enhanced polyhydroxyalkanoate (PHA) production via particulate methane monooxygenase (pMMO), the method comprising:

(a) contacting a culture of methanotrophic microorganisms with a medium comprising copper and one or more additional nutrients and a carbon-containing gas that can be metabolized by said culture;

(b) incubating said culture in said medium to cause growth of said culture; and

(c) limiting the concentration of copper in said medium;

(d) allowing or causing, for a first period of time, at least one of said one or more additional nutrient to decrease to a concentration to cause said methanotrophic microorganisms to generate PHA;

(e) causing said PHA-producing microorganisms of step (d) to grow using pMMO by exposing, for a second period of time, said PHA-producing microorganisms to the culture medium of (a); and

(f) subjecting said culture to at least one additional repetition of steps (a)-(e) resulting in a culture wherein PHA is generated by pMMO at a greater rate as compared to soluble methane monooxygenase (sMMO).

2. The method of claim 1 , wherein said culturing is performed under non-sterile conditions.

3. The method of claim 1 , wherein said culture of methanotrophic microorganisms is capable of producing intracellular PHA concentrations of at least 71% of total dry cell weight of said methanotrophic microorganisms.

4. The method of claim 1 , wherein said copper concentration is limited to be between about 0.001 micromolar and about 1000 micromolar.

5. The method of claim 1 , wherein said first period of time ranges from about 2 to 24 hours.

6. The method of claim 1 , wherein said second period of time ranges from about 12 to 24 hours.

7. The method of claim 1 , wherein said additional nutrient comprises at least one of the nutrients selected from the group consisting of aluminum, boron, calcium, carbon, carbon dioxide, cobalt, iron, magnesium, molybdenum, nitrogen, oxygen, phosphorus, potassium, sodium, and zinc.

8. The method of claim 1 , wherein the culture of methanotrophic microorganisms with enhanced PHA production through pMMO is characterized by:

(a) lack of the genetic material encoding soluble methane monooxygenase (sMMO), failure to transcribe DNA encoding sMMO or translate mRNA encoding sMMO, and/or failure to produce a functional sMMO enzyme;

(b) having the genetic material encoding the ethylmalonyl-CoA pathway, transcribing DNA encoding the ethylmalonyl-CoA pathway, and translating mRNA encoding the ethylmalonyl-CoA pathway; or

(c) the capability of producing polyhydroxyalkanoate (PHA) at intracellular PHA concentrations with a ratio of PHA to non-PHA biomass exceeding 3:1 on a dry weight basis.

9. The method of claim 1 , wherein the culture of methanotrophic microorganisms with enhanced PHA production through pMMO is characterized by:

(a) lack of the genetic material encoding soluble methane monooxygenase (sMMO), failure to transcribe DNA encoding sMMO or translate mRNA encoding sMMO, and/or failure to produce a functional sMMO enzyme;

(b) having the genetic material encoding the ethylmalonyl-CoA pathway, transcribing DNA encoding the ethylmalonyl-CoA pathway, and translating mRNA encoding the ethylmalonyl-CoA pathway; and

(c) the capability of producing polyhydroxyalkanoate (PHA) at intracellular PHA concentrations with a ratio of PHA to non-PHA biomass exceeding 3:1 on a dry weight basis.

10. The method of claim 1 , wherein the culture of methanotrophic microorganisms comprises microorganisms from a genus selected from a group consisting of: Methylosinus, Methylocystis, Methylococcus, Methylobacterium , and Pseudomonas.

11. A method for generating a culture of methanotrophic microorganisms capable to polyhydroxyalkanoate (PHA) production at intracellular PHA concentrations with a ratio of PHA to non-PHA biomass exceeding 3:1 on a dry weight basis, the method comprising:

(a) providing a culture of methanotrophic microorganisms capable of metabolizing a carbon-containing gas;

(b) providing a culture a medium comprising copper and one or more additional nutrient;

(c) providing a carbon-containing gas that can be metabolized by said culture;

(d) incubating the culture of methanotrophic microorganisms in the culture medium and in the presence of the carbon-containing gas to cause growth of said culture;

(e) limiting the concentration of copper in said medium;

(f) for a first period of time, allowing or causing at least one of said one or more additional nutrients to decrease to a concentration that results in the methanotrophic microorganisms generating PHA;

(g) causing said PHA-producing microorganisms of step (f) to grow using pMMO by exposing, for a second period of time, said PHA-producing microorganisms to the culture medium of (b) and the carbon-containing gas of (c) and incubating the culture as in step (d); and

(h) repeating steps (e)-(g) a plurality of times to result in a culture producing progressively less sMMO and progressively more pMMO, thereby yielding intracellular PHA concentrations with a ratio of PHA to non-PHA biomass exceeding 3:1 on a dry weight basis.

12. The method of claim 1 , wherein the culture of methanotrophic microorganisms comprises microorganisms from a genus selected from a group consisting of: Methylosinus, Methylocystis, Methylococcus, Methylobacterium , and Pseudomonas.

13. The method of claim 11 , wherein the limited copper concentration is between about 0.001 micromolar and about 1000 micromolar.

14. The method of claim 11 , wherein the resulting culture of methanotrophic microorganisms lacks the genetic material encoding soluble methane monooxygenase (sMMO), fails to transcribe DNA encoding sMMO or translate mRNA encoding sMMO, and/or fails to produce a functional sMMO enzyme.

15. The method of claim 11 , wherein the culture of methanotrophic microorganisms exhibits particulate methane monooxygenase activity in the presence of copper ion concentrations between 0.001 micromolar and 1000 micromolar.

16. The method of claim 11 , wherein said methanotrophic microorganisms exhibit ethylmalonyl-CoA pathway activity in the presence of copper concentrations between 0.001 micromolar and 1000 micromolar.

17. The method of claim 11 , wherein said one or more additional nutrient comprises at least one of the nutrients selected from the group consisting of aluminum, boron, calcium, carbon, carbon dioxide, cobalt, iron, magnesium, molybdenum, nitrogen, oxygen, phosphorus, potassium, sodium, and zinc.

The method of claim 11 , wherein at least one of said one or more additional nutrient comprises dissolved oxygen and wherein the method further comprises increasing the concentration of dissolved oxygen in said culture media to preferentially select for methanotrophic microorganisms exhibiting reduced pigmentation.

The method of claim 1 , wherein at least one of said one or more additional nutrient comprises dissolved oxygen and wherein the method further comprises increasing the concentration of dissolved oxygen in said culture media to preferentially select for methanotrophic microorganisms exhibiting reduced pigmentation.

20. A composition comprising isolated microorganisms obtained from the method of claim 1 .
