Abstract

Provided for herein are mutant VSV-G polypeptides, compositions comprising the same, and methods of using the same. Also provided for herein are polypeptides and compositions that bind to CD7 and uses thereof. Also provided for herein are polypeptides and compositions that bind to CD8 and uses thereof.

A polypeptide, antibody, or antibody fragment thereof, comprising a heavy chain variable domain (VH) comprising an amino sequence having at least 95% identity to SEQ ID NO: 47 and a light chain variable domain (VL) comprising an amino sequence having at least 95% identity to SEQ ID NO: 48, wherein the VH and VL are linked with a peptide linker.

72 . The polypeptide, antibody, or antibody fragment thereof, of claim 71 , comprising an antigen binding domain comprising an amino acid sequence having at least 95% identity to an amino acid sequence of SEQ ID NO: 51 or SEQ ID NO: 52.

73 . The polypeptide, antibody, or antibody fragment thereof, of claim 72 , wherein the antigen binding domain comprises an amino acid sequence having at least 95% identity to SEQ ID NO: 51.

74 . The polypeptide, antibody, or antibody fragment thereof, of claim 72 , wherein the antigen binding domain comprises an amino acid sequence having at least 95% identity to SEQ ID NO: 52.

75 . The polypeptide, antibody, or antibody fragment thereof, of claim 71 , wherein the polypeptide, antibody, or antibody fragment thereof, comprises an HCDR1 comprising an amino acid sequence of SEQ ID NO: 35, an HCDR2 comprising an amino acid sequence of SEQ ID NO: 36, an HCDR3 comprising an amino acid sequence of SEQ ID NO: 37, an LCDR1 comprising an amino acid sequence of SEQ ID NO: 38, an LCDR2 comprising an amino acid sequence of SEQ ID NO: 39, and an LCDR3 comprising an amino acid sequence of SEQ ID NO: 40.

76 . The polypeptide, antibody, or antibody fragment thereof, of claim 71 , wherein the polypeptide, antibody, or antibody fragment is an scFv.

A polypeptide, antibody, or antibody fragment thereof, comprising a heavy chain variable domain (VH) comprising an amino sequence having at least 95% identity to SEQ ID NO: 47 and a light chain variable domain (VL) comprising an amino sequence having at least 95% identity to SEQ ID NO: 48, wherein the VH and VL are linked with a peptide linker, wherein the polypeptide, antibody, or antibody fragment thereof, comprises an HCDR1 comprising an amino acid sequence of SEQ ID NO: 35, an HCDR2 comprising an amino acid sequence of SEQ ID NO: 36, an HCDR3 comprising an amino acid sequence of SEQ ID NO: 37, an LCDR1 comprising an amino acid sequence of SEQ ID NO: 38, an LCDR2 comprising an amino acid sequence of SEQ ID NO: 39, and an LCDR3 comprising an amino acid sequence of SEQ ID NO: 40.

78 . The polypeptide, antibody, or antibody fragment thereof, of claim 77 , comprising antigen binding domain comprising an amino acid sequence having at least 95% identity to an amino acid sequence of SEQ ID NO: 51 or SEQ ID NO: 52.

79 . The polypeptide, antibody, or antibody fragment thereof, of claim 77 , wherein the polypeptide, antibody, or antibody fragment thereof is an scFv.

80 . A nucleic acid molecule encoding the polypeptide, antibody, or antibody fragment thereof, of claim 71 .

81 . A vector comprising the nucleic acid molecule of claim 80 .

82 . A viral particle comprising the polypeptide, antibody, or antibody fragment thereof, of claim 71 .

83 . The viral particle of claim 82 , wherein the polypeptide, antibody, or antibody fragment thereof, is attached to the surface of the viral particle through a Fc domain.

84 . The viral particle of claim 82 , further comprising a nucleic acid molecule encoding for a heterologous molecule of interest.

85 . The viral particle of claim 84 , wherein the heterologous molecule of interest is an siRNA, an shRNA, a non-coding RNA, a polypeptide, a viral payload, a viral genome, or a combination thereof.

86 . The viral particle of claim 84 , wherein the heterologous molecule of interest is a chimeric antigen receptor (“CAR”).

87 . A cell comprising the polypeptide, antibody, or antibody fragment thereof, of claim 71 .

88 . A cell comprising the nucleic acid molecule of claim 80 .

89 . A cell comprising the viral particle of claim 82 .

90 . A method of delivering a heterologous molecule of interest to a cell, the method comprising administering the viral particle of claim 84 to the cell, wherein the cell expresses CD7.
