Abstract

Analyte collection and testing systems and methods, and more particularly to testing systems and methods that achieve significant improvements in the detection of fluorescence signals in the reader by modulating the applied optical excitation. Also described herein are optical detection apparatuses and methods for removable photonic chips that do not require translation for calibration when coupling the photonics chip with the sensing system. Also described herein are methods and apparatuses for accurately calibrating a dilution factor when reading from a photonics chip.

Claims (36)

2 . A method of calibrating a dilution factor to determine concentration of an analyte from a photonic chip, the method comprising:

measuring the binding slope of a DF calibrator material in a control solution on the photonic chip;

measuring the binding slope of the DF calibrator material in a sample solution on the photonic chip;

determining a calibrated dilution factor from the ratio of the slope of the DF calibrator material in the sample solution and the slope of the DF calibrator material in the control solution; and

outputting a concentration of the analyte based on the calibrated dilution factor.

3 . The method of claim 2 , further comprising determine the concentration of the analyte from the photonic chip using the calibrated dilution factor.

4 . The method of claim 2 , further comprising adding a dilution factor (DF) calibrator material to the control solution.

5 . The method of claim 2 , further comprising adding the DF calibrator material to the sample solution.

6 . The method of claim 2 , wherein the control solution contains a known about of the DF calibrator material.

7 . The method of claim 2 , wherein the DF calibrator material in the sample solution is diluted from a known concentration added to the sample solution.

8 . The method of claim 2 , wherein determining the calibrated dilution factor comprises comparing the ratio to a database of empirically derived calibration factors.

9 . The method of claim 2 , wherein the DF calibrator material comprises a fluorophore labeled biomolecule or microparticle.

10 . The method of claim 2 , wherein the sample solution comprises saliva.

11 . A method of calibrating a dilution factor to determine concentration of an analyte from a photonic chip, the method comprising:

adding a dilution factor (DF) calibrator material to a control solution;

measuring the binding slope of the DF calibrator material in the control solution on the photonic chip;

adding the DF calibrator material to a sample solution;

measuring the binding slope of the DF calibrator material in the sample solution on the photonic chip;

determining a calibrated dilution factor from the ratio of the slope of the DF calibrator material in the sample solution and the slope of the DF calibrator material in the control solution; and

outputting a concentration of the analyte based on the calibrated dilution factor.

12 . An optical reader device for reading a photonic chip, the device comprising:

a holder configured to secure a photonic chip to be read by the reader device;

a scan head, wherein the scan head comprises a laser light source and an optical detector; and

a controller configured to coordinate the scan head, illumination of the laser light source, and detection by the optical detector, the controller further configured to:

measure the binding slope of a DF calibrator material in a control solution on the photonic chip;

measure the binding slope of the DF calibrator material in a sample solution on the photonic chip; and

determine a calibrated dilution factor from the ratio of the slope of the DF calibrator material in the sample solution and the slope of the DF calibrator material in the control solution;

read an optical signal corresponding to an analyte from the photonic chip; and

output a concentration of the analyte from the photonic chip using the calibrated dilution factor.

13 . The device of claim 12 , wherein determining the calibrated dilution factor comprises comparing the ratio to a database of empirically derived calibration factors.

14 . The device of claim 12 , wherein the controller is configured to modulate the illumination of the laser light source with a shift signal having a frequency of 10 kHz or greater.

The device of claim 12 , wherein the controller is configured to demodulate an output signal from the optical detector using a modulated pulsed excitation signal to form a demodulated output signal, wherein the demodulated output signal is proportional to a cosine of a phase difference between the modulated signal and the output signal.

16 . The device of claim 15 , wherein the controller is configured to filter the demodulated output signal with a low-pass filter.

17 . The device of claim 15 , wherein the controller is configured to apply a matched filter to the demodulated output signal.

18 . The device of claim 15 , wherein the excitation signal has a pulse width of less than 100 ms.
