Hydrocortisone Acetate Suppository Formulation For Treatment Of Disease
The present invention relates, in various embodiments, to formulations comprising hydrocortisone and silicon dioxide. In additional embodiments, the invention relates to suppositories comprising hydrocortisone and silicon dioxide. The formulations of the present invention are useful for administration to patients who have gastrointestinal diseases and disorders such as, for example, inflammatory bowel disease.

A suppository comprising 85 mg to 95 mg hydrocortisone acetate, a suspending/dispersing agent, and an oleaginous base, wherein the suppository releases at least about 80% of the hydrocortisone acetate by about 180 minutes following exposure to dissolution media comprising a buffered 5% w/v sodium lauryl sulfate solution having a final pH in the range of about 6.8-7.0.
6 . The suppository of claim 5 , wherein the suppository has a weight of 2 grams and further comprises a polyethylene/polyvinyl chloride suppository shell.
7 . The suppository of claim 5 , wherein the suppository comprises about 90 mg hydrocortisone acetate.
8 . The suppository of claim 5 , wherein the suspending/dispersing agent is colloidal silicon dioxide or alginic acid.
9 . The suppository of claim 5 , wherein the suspending/dispersing agent is colloidal silicon dioxide.
10 . The suppository of claim 5 , wherein the oleaginous base further comprises triglycerides.
11 . The suppository of claim 5 , wherein the oleaginous base further comprises a phospholipid.
12 . The suppository of claim 11 , wherein the phospholipid is lecithin.
13 . A suppository comprising 85 mg to 95 mg hydrocortisone acetate, a suspending/dispersing agent, and an oleaginous base, wherein the suppository has a melting temperature in the range of about 35° C. to about 41° C.
14 . The suppository of claim 12 , wherein the melting temperature range is about 37° C. to about 39° C.
15 . The suppository of claim 12 , wherein the suppository has a weight of 2 grams and further comprises a polyethylene/polyvinyl chloride suppository shell.
16 . The suppository of claim 12 , wherein the suppository comprises about 90 mg hydrocortisone acetate.
17 . The suppository of claim 23 , wherein the suspending/dispersing agent is colloidal silicon dioxide or alginic acid.
18 . The suppository of claim 12 , wherein the suspending/dispersing agent is colloidal silicon dioxide.
19 . The suppository of claim 12 , wherein the oleaginous base further comprises triglycerides.
20 . The suppository of claim 12 , wherein the oleaginous base further comprises a phospholipid.
21 . The suppository of claim 20 , wherein the phospholipid is lecithin.
22 . A method of treating a gastrointestinal disease, comprising rectally administering to a subject in need thereof a suppository as claimed in claim 5 .
23 . A method of treating a gastrointestinal disease, comprising rectally administering to a subject in need thereof a suppository as claimed in claim 12 .
24 . The method of claim 22 , wherein the gastrointestinal disease is ulcerative colitis.
25 . The method of claim 22 , wherein the gastrointestinal disease is Crohn's disease.
26 . The method of claim 22 , wherein the gastrointestinal disease is hemorrhoids.
27 . The method of claim 22 , wherein the suppository is placed in a section of the rectum located between approximately 4 cm and 15 cm from the anus.
28 . The method of claim 23 , wherein the gastrointestinal disease is ulcerative colitis.
29 . The method of claim 23 , wherein the gastrointestinal disease is Crohn's disease.
30 . The method of claim 23 , wherein the gastrointestinal disease is hemorrhoids.
31 . The method of claim 23 , wherein the suppository is placed in a section of the rectum located between approximately 4 cm and 15 cm from the anus.
