Abstract

Decoding methods are provided for identifying populations in assays, particularly multiplexing assays and those associated with fluidic devices.

Claims (12)

1 . An assay decoding system, comprising:

a circuit comprising at least one processor;

a fluidic analysis chip comprising a plurality of solid supports; and

at least one of a thermal source or a light source in communication with the plurality of solid supports,

wherein the circuit activates the at least one of the thermal source or the light source to cause at least one detectable physical and/or chemical change associated with at least some of the plurality of solid supports and compares first and second encoding signals for a respective solid support in the plurality of solid supports to decode different populations of solid supports in the plurality of solid supports.

2 . The system of claim 1 , wherein the at least one of the thermal source or the light source is a photobleaching light source.

3 . The system of claim 1 , wherein the at least one of the thermal source or the light source is the thermal source.

4 . The system of claim 1 , wherein a portion of the plurality of solid supports comprises solid supports having a first dye and a second dye in different concentrations and/or ratios.

5 . The system of claim 1 , further comprising an optical detector configured to emit light at one or more defined wavelengths in communication with the circuit and the fluidic analysis chip.

6 . The system of claim 1 , wherein the at least one of the thermal source or the light source causes an irreversible, detectable physical and/or chemical change in a subset of solid supports in the plurality of solid supports for the at least one detectable physical or chemical change.

7 . The system of claim 1 , wherein at least some solid supports in the plurality of solid supports comprise first and second spectrally similar encoding agents at a first point in time, wherein one of the first and second spectrally similar encoding agents is stable and the other is unstable, and

wherein different ones of the solid supports have the first and second encoding agents in different ratios and/or concentrations to provide a plurality of different distinguishable fluorescence intensity levels before and/or after the at least one detectable physical and/or chemical change to provide a plurality of encoding states.
