Abstract

The present disclosure relates to compositions and methods that enable the formation of pharmaceutically relevant particles that can be used for therapy. In particular, the methods disclosed herein allow the controlled formation of circular particles having low internal void spaces comprising bioactive therapeutic agents.

Claims (32)

1 . A method of producing particles comprising a therapeutic biologic, the method comprising:

forming droplets of an aqueous first liquid, wherein the first liquid comprises a therapeutic biologic, and wherein the droplets are formed within a second liquid comprising an organic solvent;

dehydrating the droplets in the second liquid to form particles comprising the therapeutic biologic.

2 . The method of claim 1 , wherein the droplets are formed in a microfluidic device or homogenizer.

3 . The method of claim 2 , wherein the droplets formed in the microfluidic device are regularly spaced in the microfluidic device.

4 . The method of claim 1 , wherein dehydrating the droplets comprises contacting the second liquid with a drying substance.

5 . The method of claim 4 , wherein the drying substance is a desiccant.

6 . The method of claim 1 , further comprising drying the particles, wherein a plurality of the particles have a water content of less than 10% by weight after drying.

7 . The method of claim 6 , wherein drying the particles comprises removing the first and second liquids.

8 . The method of claim 7 , wherein the first and second liquids are removed through centrifugation, sieving, filtration, magnetic collection, solvent exchange, or decanting.

9 . The method of claim 7 , wherein residual quantities of the first and second liquids in the particles after drying are from about 0% to about 10% by weight.

10 . The method of claim 7 , wherein drying the particles further comprises contacting the particles with a stream of gas.

11 . The method of claim 10 , wherein the gas has a temperature from about −80° C. to about 200° C.

12 . The method of claim 10 , wherein the gas is helium, air, nitrogen or argon.

13 . The method of claim 7 , wherein drying the particles further comprises using lyophilization or vacuum desiccation.

14 . The method of claim 1 , further comprising removing the particles from the second liquid.

15 . The method of claim 14 , wherein the particles are removed from the second liquid through centrifugation, sieving, filtration, magnetic collection, solvent exchange, inertial separation, hydrocyclone separation, or decanting.

16 . The method of claim 6 , further comprising washing the dried particles with a washing fluid.

17 . The method of claim 16 , wherein the washing fluid is an organic liquid, a supercritical fluid, a cryogenic liquid, or a combination thereof.

18 . The method of claim 1 , wherein the therapeutic biologic is an antibody.

19 . The method of claim 18 , wherein the antibody is an IgG antibody.

20 . The method of claim 1 , wherein the therapeutic biologic in the particles has an activity per unit of about 0.5 to about 1.0.

21 . The method of claim 7 , wherein the particles comprise less than about 10% internal void spaces.

22 . The method of claim 7 , wherein the particles have less than about 10% aggregation of the therapeutic biologic.

23 . The method of claim 7 , wherein the particles have greater than about 60% therapeutic biologic by weight.

24 . The method of claim 7 , wherein the circularity of the particles is from about 0.80 to about 1.00.

25 . The method of claim 1 , wherein the first liquid is water, 0.9% saline, lactated Ringer's solution, buffers, dextrose 5%, or a combination thereof.

26 . The method of claim 1 , wherein the first liquid is water.

27 . The method of claim 1 , wherein the concentration of the therapeutic biologic in the first liquid is from about 10 mg/ml to about 500 mg/mL.

28 . The method of claim 1 , wherein the first liquid further comprises a surfactant.

29 . The method of claim 1 , wherein the organic solvent is ethyl acetate or butyl acetate.

30 . The method of claim 1 , wherein the droplets are formed in a vessel comprising tubing.
