Abstract

Disclosed is a method and an oral composition for treating CNS disorders. An embodiment of the invention comprises orally administering an allopregnanolone containing composition to a subject having a CNS disorder. Exemplary CNS disorders include sleep disorders, mood disorders, dysthymic disorders, bipolar disorders, anxiety disorders, stress disorders, compulsive disorders, schizophrenia spectrum disorders, convulsive disorders, memory disorders, cognition disorders, movement disorders, personality disorders, autism spectrum disorders, substance abuse disorders, dementia, pain, traumatic brain injury (TBI), vascular disease, withdrawal syndrome, and tinnitus. The composition and methods disclosed herein exhibit effective oral absorption/bioavailability.

Claims (61)

1. A method of treating a PPD in a subject, said method comprising orally administering to said subject a tablet form oral pharmaceutical composition (PC) and fat containing food,

wherein said subject comprises at least one of an adolescent human female, and an adult human female, and

wherein said PC comprises allopregnanolone in an amount of from about 200 mg to about 300 mg, and

wherein said administration comprises a dosing regimen, said dosing regimen comprising a plurality of doses, and

wherein said regimen comprises a minimum daily dosing of three doses, and

wherein said dose comprises at least two tablet forms, and

wherein said regimen comprises a minimum daily allopregnanolone administration of from about 1400 mg to about 1600 mg, and

wherein said regimen comprises a minimum duration of at least one of 48 hours, and

wherein said administration of said PC to said subject results in a C max of at least 10 ng/mL of allopregnanolone in said subject.

2. The method of claim 1 , wherein said PC comprises at least one of a hydrophilic additive and a lipophilic additive.

3. The method of claim 1 , wherein said PC comprises a plurality of additives, said plurality of additives comprising at least one surfactant.

4. The method of claim 1 , wherein said C max comprises at least one of at least 15 ng/mL, at least 20 ng/mL, at least 25 ng/mL, at least 30 ng/mL, at least 35 ng/mL, and at least 40 ng/mL.

5. The method of claim 1 , wherein said C max occurs after at least two days of administration of said PC.

6. The method of claim 1 , wherein said regimen comprises at least one of a titrated dose, an untitrated dose, and a hybrid thereof.

7. The method of claim 1 , wherein in response to said administration of said composition to said subject, said PPD of said subject is at least one of substantially improved, reduced, and eliminated.

8. A method of treating a depression in a subject, said method comprising orally administering to said subject a tablet form oral pharmaceutical composition (PC) and fat containing food,

wherein said subject comprises at least one of an adolescent human male, an adult human male, an adolescent human female, and an adult human female, and

wherein said PC comprises allopregnanolone in an amount of from about 150 mg to about 350 mg, and

wherein said administration comprises a dosing regimen, said dosing regimen comprising a plurality of doses, and

wherein said regimen comprises a minimum daily dosing of a plurality of doses, and

wherein said dose comprises a plurality of tablet forms, and

wherein said regimen comprises a minimum daily allopregnanolone administration of from about 1200 mg to about 1800 mg, and

wherein said regimen comprises a minimum duration of at least one of 24 hours, 48 hours, and more than 48 hours, and

wherein said administration of said PC to said subject results in a C max of at least 10 ng/mL of allopregnanolone in said subject.

9. The method of claim 8 , wherein said PC comprises at least one of a hydrophilic additive and a lipophilic additive.

10. The method of claim 8 , wherein said PC comprises a plurality of additives, said plurality of additives comprising at least one surfactant.

11. The method of claim 8 , wherein said C max comprises at least one of at least 15 ng/mL, at least 20 ng/mL, at least 25 ng/mL, at least 30 ng/mL, at least 35 ng/mL, and at least 40 ng/mL.

12. The method of claim 8 , wherein said C max occurs after at least two days of administration of said PC.

13. The method of claim 8 , wherein said regimen comprises at least one of a titrated dose, an untitrated dose, and a hybrid thereof.

14. The method of claim 8 , wherein in response to said administration of said composition to said subject, said depression of said subject is at least one of substantially improved, reduced, and eliminated.

The method of claim 8 , wherein said depression comprises at least one of clinical depression, postnatal depression, postpartum depression, perinatal depression, atypical depression, melancholic depression, psychotic major depression, catatonic depression, double depression, depressive personality disorder, recurrent brief depression, minor depressive disorder, manic depressive disorder, chronic medical condition depression, treatment-resistant depression, and refractory depression.

16. The method of claim 8 , wherein said PC comprises allopregnanolone in an amount of from about 200 mg to about 300 mg, and wherein said regimen comprises a minimum daily allopregnanolone administration of from about 1400 mg to about 1600 mg.

17. The method of claim 8 , wherein said minimum daily dosing comprises three doses, and wherein said plurality of tablet forms comprises two tablet forms.

18. A method of treating a depression in a subject, said method comprising orally administering to said subject a tablet form oral pharmaceutical composition (PC),

wherein said subject comprises at least one of an adolescent human male, an adult human male, an adolescent human female, and an adult human female, and

wherein said PC comprises allopregnanolone in an amount of from about 100 mg to about 400 mg, and

wherein said administration comprises a dosing regimen, said dosing regimen comprising a plurality of doses, and

wherein said dose comprises a plurality of tablet forms, and

wherein said regimen comprises a minimum daily allopregnanolone administration of from about 1200 mg to about 1800 mg, and

wherein said regimen comprises a minimum duration of at least one of 24 hours, 48 hours, and more than 48 hours, and

wherein said administration of said PC to said subject results in a C max of at least 10 ng/mL of allopregnanolone in said subject.

19. The method of claim 18 , wherein said PC comprises at least one of a hydrophilic additive and a lipophilic additive.

20. The method of claim 18 , wherein said PC comprises a plurality of additives, said plurality of additives comprising at least one surfactant.

21. The method of claim 18 , wherein said C max comprises at least one of at least 15 ng/mL, at least 20 ng/mL, at least 25 ng/mL, at least 30 ng/mL, at least 35 ng/mL, and at least 40 ng/mL.

22. The method of claim 18 , wherein said C max occurs after at least two days of administration of said PC.

23. The method of claim 18 , wherein said regimen comprises at least one of a titrated dose, an untitrated dose, and a hybrid thereof.

24. The method of claim 18 , wherein in response to said administration of said composition to said subject, said depression of said subject is at least one of substantially improved, reduced, and eliminated.

25. The method of claim 18 , wherein said depression comprises at least one of clinical depression, postnatal depression, postpartum depression, perinatal depression, atypical depression, melancholic depression, psychotic major depression, catatonic depression, double depression, depressive personality disorder, recurrent brief depression, minor depressive disorder, manic depressive disorder, chronic medical condition depression, treatment-resistant depression, and refractory depression.

26. The method of claim 18 , wherein said PC comprises allopregnanolone in an amount of from about 200 mg to about 300 mg, and wherein said regimen comprises a minimum daily allopregnanolone administration of from about 1400 mg to about 1600 mg.

27. The method of claim 18 , wherein said minimum daily dosing comprises three doses, and wherein said plurality of tablet forms comprises two tablet forms.

28. The method of claim 18 , wherein said administration comprises administering fat containing food.

29. The method of claim 18 , wherein said C max is obtaining regardless of a fed/fasted state of said subject.

30. A method of treating a depression in a subject, said method comprising orally administering to said subject a tablet form oral pharmaceutical composition (PC),

wherein said subject comprises at least one of an adolescent human male, an adult human male, an adolescent human female, and an adult human female, and

wherein said PC comprises allopregnanolone in an amount of from about 200 mg to about 300 mg, and

wherein said administration comprises a dosing regimen, said dosing regimen comprising a plurality of doses, and

wherein said regimen comprises a minimum daily dosing of three doses, and

wherein said dose comprises at least two tablet forms, and

wherein said regimen comprises a minimum daily allopregnanolone administration of from about 1400 mg to about 1600 mg, and

wherein said regimen comprises a minimum duration of at least one of 48 hours, and

wherein said administration of said PC to said subject results in a C max of at least 10 ng/mL of allopregnanolone in said subject.
