Abstract

Methods and systems are provided for generating and utilizing a genetically engineered bacterium comprising a modification in glnD, wherein said modification is selected from the group consisting of: deletion of the entire gene, deletion of substantially the entire gene, deletion of an ACT domain, deletion of more than 50% of an ACT domain, deactivation of an ACT domain, and deactivation of an UTase domain.

48 . A genetically engineered diazotrophic bacterium having decreased expression of GlnA, wherein: GlnA expression is decreased by replacing the native promoter of glnA with a promoter with lower activity.

49 . The genetically engineered diazotrophic bacterium of claim 48 , wherein the native promoter of glnA is replaced with a promoter selected from the group consisting of: a glnB promoter, a glnD promoter, and a glnE promoter.

50 . The genetically engineered diazotrophic bacterium of claim 48 , wherein the genetically engineered diazotrophic bacterium is intergeneric.

51 . The genetically engineered diazotrophic bacterium of claim 48 , wherein the genetically engineered diazotrophic bacterium is capable of fixing atmospheric nitrogen in the presence of exogenous nitrogen.

52 . The genetically engineered diazotrophic bacterium of claim 48 , wherein the genetically engineered diazotrophic bacterium comprises an additional genetic modification in a nitrogen fixation genetic network or a nitrogen assimilation genetic network.

53 . The genetically engineered diazotrophic bacterium of claim 52 , wherein the additional genetic modification comprises a genetic modification that results in one or more of increased expression of NifA, decreased expression of NifL, and increased expression of NifHl.

54 . The genetically engineered diazotrophic bacterium of claim 52 , wherein the additional genetic modification comprises a genetic modification that results in increased expression of NifA and decreased expression of NifL.

55 . The genetically engineered diazotrophic bacterium of claim 52 , wherein the additional genetic modification comprises a genetic modification in nfL.

56 . The genetically engineered diazotrophic bacterium of claim 52 , wherein the additional genetic modification comprises a genetic modification in glnE.

57 . The genetically engineered diazotrophic bacterium of claim 52 , wherein the additional genetic modification comprises a genetic modification that results in decreased activity of GlnE.

58 . A method of increasing the amount of atmospheric derived nitrogen in a plant, comprising contacting the plant with a plurality of the genetically engineered diazotrophic bacterium of claim 48 .

59 . The method of claim 58 , wherein the plurality of genetically engineered diazotrophic bacteria is applied to a seed of the plant, a seedling of the plant, a plant after planting but before harvest of the plant, or a plant as a side dressing.

60 . The method of claim 58 , wherein the plurality of genetically engineered diazotrophic bacteria is applied to the plant in a liquid formulation.

61 . The method of claim 58 , wherein the plurality of genetically engineered diazotrophic bacteria is applied to the plant between one month and eight months, between two months and eight months, between one month and three months, or between three months and six months after germination.

62 . The method of claim 58 , wherein the plant is a cereal plant, a corn plant, a rice plant, a wheat plant, or a soy plant.

63 . A composition comprising a seed, and a seed coating; wherein the seed coating comprises a plurality of the genetically engineered diazotrophic bacterium of claim 48 .

64 . The composition of claim 63 , wherein the seed is a cereal seed, a corn seed, a wheat seed, a rice seed, a soy seed, a rye seed, or a sorghum seed.

65 . A composition comprising a plant and a plurality of the genetically engineered diazotrophic bacterium of claim 48 .

66 . The composition of claim 65 , wherein the plant is a seedling.

67 . The composition of claim 65 , wherein the plant is a cereal plant, a corn plant, a rice plant, a wheat plant, a soy plant, a rye plant, or a sorghum plant.
