Abstract

The invention generally relates to plant cells and plants modified to increase resistance to necrotrophs or drought and methods of selecting and using the same. More specifically, the invention relates in part to plant cells and/or plants modified to eliminate or reduce as compared to control plants cell the NADPH oxidase activity or expression of certain respiratory burst oxidase homolog (RBOH) proteins and methods of selecting for and using the same.

Claims (20)

30 . A plant cell transformed with a first expression cassette comprising a heterologous promoter operably linked to a sequence encoding a first polynucleotide; wherein expression of said first polynucleotide in said plant cell reduces or eliminates expression of at least one endogenous respiratory burst oxidase protein, increases resistance to a necrotroph, and increases drought tolerance in said plant cell as compared to a control plant cell of the same species lacking said first expression cassette; wherein said at least one endogenous respiratory burst oxidase protein has at least 97% sequence identity to SEQ ID NO: 3 or SEQ ID NO: 4; and wherein the plant cell is selected from the group consisting of a soybean plant cell, a common bean plant cell, and a leguminous plant cell.

31 . The transformed plant cell of claim 1 , wherein expression of at least two endogenous respiratory burst oxidase proteins is reduced or eliminated.

32 . The transformed plant cell of claim 1 , wherein expression of at least four endogenous respiratory burst oxidase proteins is reduced or eliminated.

33 . The transformed plant cell of claim 1 , wherein said at least one endogenous respiratory burst oxidase protein has at least 98% sequence identity to SEQ ID NO: 3 or SEQ ID NO: 4.

34 . The transformed plant cell of claim 1 , wherein said at least one endogenous respiratory burst oxidase protein has at least 99% sequence identity to SEQ ID NO: 3 or SEQ ID NO: 4.

35 . The transformed plant cell of claim 1 , wherein said at least one endogenous respiratory burst oxidase protein has the amino acid sequence set forth in SEQ ID NO: 3 or the amino acid sequence set forth in SEQ ID NO: 4.

36 . The transformed plant cell of claim 1 , wherein expression of said at least one endogenous respiratory burst oxidase protein is reduced by at least 30% as compared to said control plant cell.

37 . The transformed plant cell of claim 1 , wherein the necrotroph is Sclerotinia sclerotiorum.

The transformed plant cell of claim 1 , wherein the transformed plant cell is further transformed with a second expression cassette comprising a heterologous promoter operably linked to a sequence encoding a second polynucleotide, wherein expression of said second polynucleotide in said plant cell reduces or eliminates expression of at least one additional endogenous respiratory burst oxidase protein having at least 95% sequence identity to SEQ ID NO: 3 or SEQ ID NO: 4.

39 . The transformed plant cell of claim 38 , wherein the expression of at least two additional endogenous respiratory burst oxidase proteins is reduced or eliminated.

40 . The transformed plant cell of claim 1 , wherein said first polynucleotide is selected from the group consisting of miRNA, siRNA and antisense RNA.

41 . A transgenic plant comprising the transformed plant cell of claim 1 .

42 . A transgenic plant seed obtained from the transgenic plant of claim 40 , wherein said transgenic plant seed comprises said first expression cassette.

43 . A method of generating a transgenic plant comprising:

a) transforming plant cells with a first expression cassette comprising a heterologous promoter operably linked to a sequence encoding a first polynucleotide, wherein expression of said first polynucleotide in said plant cells reduces or eliminates expression of at least one endogenous respiratory burst oxidase protein in said plant cells as compared to a control plant cell of the same species lacking said first expression cassette, wherein said at least one endogenous respiratory burst oxidase protein has at least 97% amino acid sequence identity to SEQ ID NO: 3 or SEQ ID NO: 4, and wherein said plant cells are selected from the group consisting of soybean plant cells, common bean plant cells and leguminous plant cells;

b) obtaining transgenic plants from said transformed plant cells of step (a); and

c) selecting a transgenic plant from the transgenic plants of step (b) that expresses said first polynucleotide and exhibits increased resistance to a necrotroph and increased drought tolerance as compared to a control plant of the same species lacking said first expression cassette and grown under identical growth conditions.

44 . The method of claim 43 , further comprising obtaining transgenic plant seeds from the transgenic plant selected in step (c).

The method of claim 43 , wherein the transformed plant cells generated in step (a) are further transformed with a second expression cassette comprising a heterologous promoter operably linked to a sequence encoding a second polynucleotide prior to step (b), wherein expression of said second polynucleotide in said plant cell reduces or eliminates expression of at least one additional endogenous respiratory burst oxidase protein having at least 95 % sequence identity to SEQ ID NO: 3 or SEQ ID NO: 4.

46 . The method of claim 43 , wherein said first polynucleotide is selected from the group consisting of miRNA, siRNA, and antisense RNA.
