Abstract

Methods for automated ICSI include holding an oocyte in a holding device, moving an injection pipette, and positioning the oocyte to the motor position associated with the image in which the oocyte is most in focus. The methods also include using AI to: detect and determine the thickness of a zona pellucida of an oocyte, determine an image in which the oocyte is most in focus, align the oocyte and the injection pipette, label morphological structure(s) of the oocyte, and create a trajectory for the injection pipette. The methods also include perforating (by laser or piezo) the zona pellucida and injecting a spermatozoon along the injection trajectory into the oocyte.

Claims (19)

1 . A method for automated, artificial-intelligence-based intracytoplasmic sperm injection (ICSI), the method comprising:

a) receiving an oocyte;

b) using an artificial intelligence system and an imaging device to detect a zona pellucida, wherein the imaging device includes microscope and a camera; and

c) holding the oocyte in a holding device;

d) moving an injection pipette;

e) using the artificial intelligence system and the imaging device to determine an image in which the oocyte is most in focus;

f) using the artificial intelligence system to align the oocyte and the injection pipette;

g) using the artificial intelligence system to label one or more morphological structures of the oocyte;

h) positioning the oocyte to a motor position associated with the image in which the oocyte is most in focus;

i) using the artificial intelligence system and the imaging device to detect a thickness of the zona pellucida;

j) perforating, by a laser or a piezo, the zona pellucida;

k) using the artificial intelligence system and the imaging device to create an injection trajectory for the injection pipette; and

l) injecting a spermatozoon along the injection trajectory into the oocyte.

2 . The method of claim 1 , wherein the holding device is a motorized holding pipette.

3 . The method of claim 1 , wherein perforating is performed by laser.

4 . The method of claim 1 , wherein perforating is performed by piezo.

5 . The method of claim 1 , further comprising executing the injection trajectory by moving the injection pipette.

6 . The method of claim 1 , further comprising calculating where the end of the pipette tip ends up for release of the spermatozoa into the oocyte.

7 . The method of claim 1 , further comprising detecting release of the spermatozoon from the injection pipette.
