Abstract

Analyte collection and testing systems and methods, and more particularly to disposable oral fluid collection and testing systems and methods. Described herein are methods and apparatuses to achieve significant improvements in the detection of fluorescence signals in the reader.

Claims (36)

1 . An optical reader device for analyte testing, the device comprising:

a cartridge holder configured to receive a removable cartridge containing a photonic chip;

a scan head comprising a first plurality of optical fibers optically connected to one or more laser light sources and a second plurality of optical fibers optically connected to a plurality of detectors;

a scan head actuator configured to move the scan head relative to the cartridge holder;

a pump membrane actuator configured to apply force to a membrane pump of the cartridge;

a controller configured to coordinate movement of the scan head, illumination of the laser light sources, detection by the detectors, and actuation of the pump membrane.

2 . The device of claim 1 , wherein the scan head comprises polarization-maintaining optical fibers configured to maintain transverse-magnetic (TM) polarization.

3 . The device of claim 1 , wherein the cartridge holder includes a ball plunger and a movable clamp base for precise alignment of the cartridge in x, y, and z axes.

4 . The device of claim 1 , further comprising one or more fluid sensors configured to detect fluid presence within the cartridge.

5 . The device of claim 1 , wherein the pump membrane actuator comprises a rocker arm with a rounded end for uniform force application.

6 . The device of claim 1 , further comprising a blister pack actuator configured to rupture a blister pack within the cartridge.

7 . The device of claim 1 , wherein the controller is configured to regulate temperature of the cartridge via integrated heaters and temperature sensors.

8 . A disposable cartridge for use with an optical reader device, the cartridge comprising:

a saliva collection system including one or more swabs;

a fluidic circuit comprising a diluted sample cavity, sample metering well, mixing chamber, and sensing chip;

a blister pack containing control fluid;

one or more capillary stops and vent ports configured to regulate fluid movement;

a photonic chip comprising excitation and emission waveguides aligned for evanescent signal detection.

9 . The cartridge of claim 8 , wherein the photonic chip includes a plurality of sensing wells coated with analyte-specific antigens.

10 . The cartridge of claim 8 , wherein the mixing chamber comprises a serpentine channel configured to enhance chaotic advection.

11 . The cartridge of claim 8 , wherein the blister pack contains a control fluid with detectably labeled antibodies.

12 . The cartridge of claim 8 , wherein the fluidic circuit includes vent ports controlled by the reader to cleave and meter fluid slugs.

13 . The cartridge of claim 8 , wherein the sensing chip is configured to detect fluorescence signals via evanescent coupling.

14 . A method for detecting an analyte in a bodily fluid sample using an optical reader device, the method comprising:

collecting a saliva sample using a swab;

mixing the sample with a dilution buffer to form a diluted sample;

transferring the diluted sample to a photonic chip;

illuminating the chip with TM-polarized light via a scan head;

detecting fluorescence signals emitted from the chip;

analyzing the signals to determine a presence or concentration of the analyte.

15 . The method of claim 14 , wherein the dilution buffer comprises bovine serum albumin (BSA) in phosphate-buffered saline (PBS).

16 . The method of claim 14 , further comprising incubating the sample in a mixing chamber prior to detection.

17 . The method of claim 14 , wherein the fluorescence signal is detected from a plurality of sensing wells and analyzed over time.

18 . The method of claim 14 , further comprising comparing a sample signal to a control signal to determine analyte concentration.

19 . The method of claim 14 , wherein the scan head is aligned with the photonic chip using a scan head actuator controlled by the reader.

20 . The method of claim 14 , wherein the reader outputs a result indicating “Pass,” “Fail,” or “Inconclusive” based on analyte detection.
