Peptides for Treating Disease and Reducing Symptoms Related to Multiple Sclerosis
Methods and materials for treating and preventing autoimmune diseases, in particular, multiple sclerosis (MS) including small peptides are capable of interacting with CD40, thereby altering and/or modulating the ability of CD40 to interact with CD154, which apparently affects inflammation; and/or the use of such peptides in reducing the inflammatory response, and in particular, the autoimmune inflammatory response; and/or the use of such short peptides to prevent or reverse autoimmune disease, and particularly, multiple sclerosis, in individuals; and/or methods and materials for detecting T-cells that express CD40 (Th40 cells). Also provided are kits for reducing inflammation, treating autoimmune diseases, or detecting Th40 cells. Additionally, methods and apparatuses to administer the peptide are provided.

50 . A peptide that treats, abates the progression of, or reverses the symptoms of multiple sclerosis;
the peptide affecting the interaction of CD40 with CD154/gp39/CD40-ligand;
the peptide being configured for administration to a subject, a therapeutically effective amount thereof that abates the progression of or reverses the symptoms of the multiple sclerosis;
the administration of the peptide being upon the subject being diagnosed with pre-MS or MS.
51 . The peptide of claim 50 , wherein the peptide binds to CD40.
52 . The peptide of claim 50 , wherein the peptide binds to a CD40 protein with a Kd of less than 10 −6 .
53 . The peptide of claim 50 , wherein the peptide binds CD40 at the site where CD40 interacts with CD154.
54 . The peptide of claim 50 , wherein the peptide blocks or inhibits the binding of CD40 to CD154.
55 . The peptide of claim 50 , wherein the peptide affects the interaction of CD40 with CD154 to alter the cytokine expression profile of a cell population treated with said peptide.
56 . The peptide of claim 50 , wherein the peptide has 10 amino acids or less in length.
57 . The peptide of claim 50 , wherein the peptide comprising an amino acid sequence selected from the group of SEQ ID NO:3, SEQ ID NO:4, SEQ ID NO:5, SEQ ID NO:6, SEQ ID NO:7, SEQ ID NO:8, SEQ ID NO:9, SEQ ID NO: 27, SEQ ID NO: 28, SEQ ID NO: 29, and SEQ ID NO: 30.
58 . The peptide of claim 50 , wherein the peptide comprising an amino acid sequence selected from the group of SEQ ID NO:4, SEQ ID NO: 27, SEQ ID NO: 28, SEQ ID NO: 29, and SEQ ID NO: 30.
59 . The peptide of claim 50 , wherein the peptide is a 6 amino acid long peptide having the amino acid sequence of SEQ ID NO: 28.
60 . The peptide of claim 50 , wherein the peptide is a 6 amino acid long peptide having the amino acid sequence of SEQ ID NO: 29.
61 . The peptide of claim 50 , wherein the peptide is configured to have one or more modifications selected from:
(a) pegylation, glycosylation, acetylation, and amidation;
(b) PASylation, or including polyglycerols, polyoxazolines, poly(amino acids), polyacrylamides, polyvinylpyrrolidones, and polyzwitterons;
(c) synthetization; and
(d) being configured as a salt.
62 . The peptide of claim 50 , wherein the multiple sclerosis is primary progressive MS (PPMS) or secondary progressive MS (SPMS)
63 . The peptide of claim 50 , wherein the administration of the peptide to the subject is before the onset and/or at the onset of symptoms of multiple sclerosis.
64 . The peptide of claim 50 , wherein the administration of the peptide alleviates symptoms of relapsing or remitting multiple sclerosis (RRMS).
65 . The peptide of claim 50 , wherein the administration of the peptide:
(a) slows or abates the growth of number of lesions;
(b) slows or abates the size of lesions; and/or
(c) prolongs tissue retention of T cells.
66 . The peptide of claim 50 , wherein the peptide modulates CD69 levels and/or IL-10 levels in the subject.
67 . The peptide of claim 50 , wherein the peptide modulates CD69 levels and/or IL-10 of draining lymph nodes and/or spleen of the subject.
68 . The peptide of claim 50 , wherein the peptide does not affect CD44 + /CD62L − levels of the subject.
69 . The peptide of claim 50 , wherein the peptide binds to Th40 cells.
