Abstract

Transgenic INIR6 maize plants comprising modifications of the DP-4114 maize locus which provide for facile excision of the modified DP-4114 transgenic locus or portions thereof, methods of making such plants, and use of such plants to facilitate breeding are disclosed.

Claims (23)

23 : A transgenic maize plant cell comprising a modified transgenic locus comprising a deletion of one or more elements of an original transgenic locus; wherein the deletion comprises a deletion of part or all of a phosphinothricin acetyltransferase (PAT) selectable marker cassette and a 3′ junction polynucleotide.

24 : The transgenic maize plant cell of claim 23 , wherein the original transgenic locus is a DP-4114 transgenic locus set forth in SEQ ID NO: 1.

25 : The transgenic maize plant cell of claim 23 , wherein the deletion comprises at least the region corresponding to nucleotides 12,734 to 14,370 of SEQ ID NO: 1; or part of the region corresponding to nucleotides 12,734 to 14,370 of SEQ ID NO: 1.

26 : The transgenic maize plant cell of claim 23 , wherein the modified transgenic locus comprises SEQ ID NO: 25 or an allelic variant thereof.

27 : The transgenic maize plant cell of claim 23 , wherein the deletion comprises a deletion of all of the phosphinothricin acetyltransferase (PAT) selectable marker cassette and part of the 3′ junction polynucleotide.

28 : The transgenic maize plant cell of claim 27 , wherein the deletion of part of the 3′ junction polynucleotide comprises between about 8 to about 1000 base pairs of endogenous chromosomal DNA of the plant genome.

29 : The transgenic maize plant cell of claim 28 , wherein the deletion of part of the 3′ junction polynucleotide comprises between about 10 to 100 base pairs of endogenous chromosomal DNA of the plant genome.

30 : A transgenic maize plant part comprising the maize plant cell of claim 23 , wherein the maize plant part optionally comprises a seed.

31 : A transgenic maize plant comprising the maize plant cell of claim 23 .

32 : A DNA molecule comprising a modified transgenic locus comprising a deletion of one or more elements of an original transgenic locus; wherein the deletion comprises a deletion of part or all of a phosphinothricin acetyltransferase (PAT) selectable marker cassette and a 3′ junction polynucleotide.

33 : The DNA molecule of claim 32 , wherein the original modified transgenic locus is a DP-4114 transgenic locus set forth in SEQ ID NO: 1.

34 : The DNA molecule of claim 32 , wherein the deletion comprises a deletion of all of the phosphinothricin acetyltransferase (PAT) selectable marker cassette and part of the 3′ junction polynucleotide.

35 : The DNA molecule of claim 34 , wherein the deletion of part of the 3′ junction polynucleotide comprises between about 8 to about 1000 base pairs of endogenous chromosomal DNA of the plant genome.

36 : The DNA molecule of claim 32 , wherein the deletion of part of the 3′ junction polynucleotide comprises between about 10 to 100 base pairs of endogenous chromosomal DNA of the plant genome.

37 : A processed transgenic maize plant product comprising the DNA molecule of claim 32 .

38 : A biological sample containing the DNA molecule of claim 32 .

39 : A nucleic acid molecule adapted for detection of genomic DNA comprising the DNA molecule of claim 32 , wherein said nucleic acid molecule optionally comprises a detectable label.

40 : A processed transgenic maize plant product comprising:

a DNA molecule comprising a modified transgenic locus comprising a deletion of one or more elements of an original transgenic locus; wherein the deletion comprises a deletion of part or all of a phosphinothricin acetyltransferase (PAT) selectable marker cassette and a 3′ junction polynucleotide; or

transgenic maize plant cell comprising a modified transgenic locus comprising a deletion of one or more elements of an original transgenic locus; wherein the deletion comprises a deletion of part or all of a phosphinothricin acetyltransferase (PAT) selectable marker cassette and a 3′ junction polynucleotide.

41 : A method of detecting the DNA molecule of claim 40 or a DNA molecule in the transgenic maize plant cell of claim 40 , comprising the step of detecting a DNA molecule comprising SEQ ID NO: 25 or an allelic variant thereof.

42 : The method of claim 40 , wherein the method comprises detecting a unique transgenic locus excision site in the processed transgenic maize plant product.
