Abstract

The invention provides for compounds of formula I wherein the substitutents are as described herein. Further provided are methods of using such compounds for the treatment of eating disorders, metabolic disorders, obesity, cognitive disorders, neurological disorders, pain disorders, inflammation disorders, in the promotion of smoking cessation and for the treatment of other psychiatric disorders Also provided are pharmaceutical compositions containing such compounds and pharmaceutical combinations of the compounds of the invention with other therapeutic agents.

Claims (35)

1. A compound of formula I

including all pharmaceutically acceptable salts and stereoisomers,

wherein:

R 1 , R 3 and R 4 are each independently selected from the group consisting of hydrogen, alkyl, halo and CN;

R 2 is selected from the group consisting of alkenyl, alkynyl, cycloalkyl, cycloalkylalkyl, heterocyclyl, heterocycloalkyl, heteroaryl, arylalkyl, heteroarylalkyl, acyl, OR 11 and OCF 2 H;

R 5 is selected from the group consisting of alkyl, alkenyl, alkynyl, cycloalkyl, heterocyclyl, aryl, heteroaryl, COOR 13 and CONR 13 R 13a ;

R 7 and R 7a are each independently selected from the group consisting of hydrogen, alkyl and cycloalkyl;

R 9 is selected from the group consisting of hydrogen, alkyl, alkenyl, alkynyl, cycloalkyl, cycloalkylalkyl, heterocyclyl, heterocycloalkyl, arylalkyl and heteroarylalkyl;

R 10 is selected from the group consisting of alkenyl, alkynyl, cycloalkyl, cycloalkylalkyl, heterocyclyl, heterocycloalkyl, aryl, heteroaryl, arylalkyl and heteroarylalkyl;

R 11 is selected from the group consisting of aryl, heteroaryl and heteroarylalkyl;

R 13 and R 13a are each independently selected from the group consisting of hydrogen, alkyl, alkenyl, alkynyl, cycloalkyl, cycloalkylalkyl, heterocyclyl, heterocycloalkyl, aryl, heteroaryl, arylalkyl and heteroarylalkyl;

or R 13 and R 13a taken together can form cycloalkyl or heterocyclyl;

X is —(CR 14 R 14a ) n —;

Y is independently selected from the group consisting of —S(O) 2 — and —SO 2 N(R 15 )—;

R 14 and R 14a are each independently selected from the group consisting of hydrogen and alkyl;

R 15 is selected from the group consisting of hydrogen, alkyl, alkenyl, alkynyl, cycloalkyl, cycloalkylalkyl, heterocyclyl, heterocycloalkyl, aryl, heteroaryl, arylalkyl and heteroarylalkyl;

or R 10 and R 15 taken together can form cycloalkyl or heterocyclyl; and

n is an integer 0, 1 or 2, with the following provisos

R 5 is neither imidazole nor substituted imidazole;

when Y is —S(O) 2 —, R 10 is not a seven-membered lactam; and

when Y is —SO 2 N(R 15 )—, neither R 10 nor R 15 is a seven-membered lactam.

2. The compound of claim 1 , including all pharmaceutically acceptable salts and stereoisomers, wherein:

R 2 is selected from the group consisting of heteroaryl, arylalkyl, OR 11 and OCF 2 H;

R 5 is selected from the group consisting of aryl and heteroaryl;

R 9 is hydrogen;

R 10 is selected from the group consisting of aryl, heteroaryl, arylalkyl and heteroarylalkyl; and

X is CH 2 .

3. The compound of claim 2 , including all pharmaceutically acceptable salts and stereoisomers, wherein:

R 1 , R 3 and R 4 are each hydrogen;

R 5 is aryl;

R 7 and R 7a are each hydrogen; and

Y is —S(O) 2 —.

4. The compound of claim 1 , including all pharmaceutically acceptable salts and stereoisomers, selected from the group consisting of:

N-(1,6-Dibenzyl-2-oxo-1,2,3,4-tetrahydroquinolin-3-yl)-benzenesulfonamide and

N-(1-Benzyl-2-oxo-6-(thiophen-3-yl)-1,2,3,4-tetrahydroquinolin-3-yl)-benzenesulfonamide.
