Methods of modulating IFNy production
Provided are cytokines and methods of modulating activity of the immune system using cytokine agonists and antagonists. Also provided are methods of treatment of immune and proliferative disorders.
1. A method of inhibiting IFNγ production from a cell, comprising contacting the cell with an effective amount of a first antibody or antibody fragment thereof that antagonizes IL-27 activity in conjunction with an effective amount of a second antibody or antibody fragment that inhibits activity of IL-12.
2. The method of claim 1 wherein the cell is a human cell selected from the group consisting of an NK cell and a T cell.
3. The method of claim 1 , wherein the first antibody or antibody fragment thereof binds to a polypeptide selected from the group consisting of p28, EBI3, a complex of p28 and EBI3, and WSX-1/TCCR.
4. The method of claim 3 , wherein the first antibody or antibody fragment thereof binds the polypeptide with a K D of at least 10 −3 M.
5. The method of claim 3 , wherein the first antibody or antibody fragment thereof binds the polypeptide with a K D of at least 10 −6 M.
6. The method of claim 3 , wherein the first antibody or antibody fragment thereof binds the polypeptide with a K D of at least 10 −11 M.
7. The method of claim 1 wherein the first and second antibody or antibody fragment thereof are humanized or chimeric.
