Abstract

Isolated human monoclonal antibodies which specifically bind to human EGFR, and related antibody-based compositions and molecules, are disclosed. The human antibodies can be produced by a transfectoma or in a non-human transgenic animal, e.g., a transgenic mouse, capable of producing multiple isotypes of human monoclonal antibodies by undergoing V-D-J recombination and isotype switching. Also disclosed are pharmaceutical compositions comprising the human antibodies, non-human transgenic animals and hybridomas which produce the human antibodies, and therapeutic and diagnostic methods for using the human antibodies.

Claims (14)

1. An isolated monoclonal antibody which binds to human EGFR, comprising a heavy and light chain variable region, wherein the heavy chain variable region comprises the amino acid sequence set forth in SEQ ID NO: 2.

2. An isolated monoclonal antibody which binds to human EGFR, comprising a heavy and light chain variable region, wherein the light chain variable region comprises the amino acid sequence set forth in SEQ ID NO: 4.

3. An isolated monoclonal antibody which binds an epitope on human EGFR recognized by an antibody comprising heavy and light chain variable regions comprising the amino acid sequences set forth in SEQ ID NOs: 2 and 4, respectively.

4. An isolated human monoclonal antibody which binds to human EGFR, competes with antibody 225, but does not compete with antibody 528.

5. The antibody of claim 4 , wherein the antibody competes for binding to human EGFR with an antibody comprising heavy and light chain variable regions comprising the amino acid sequences set forth in SEQ ID NOs: 2 and 4, respectively.

6. The antibody of any one of claims 1 - 5 , wherein the antibody is a human IgGl antibody.

7. The antibody of any one of claims 1 - 5 , wherein the antibody is an Fab fragment or a single chain antibody.

8. The antibody of any one of claims 1 - 5 produced by a hybridoma which includes a B cell obtained from a transgenic non-human animal having a genome comprising a human heavy chain transgene and a human light chain transgene fused to an immortalized cell.

9. The antibody of any one of claims 1 - 5 produced by a transfectoma comprising nucleic acids encoding a human heavy chain and a human light chain.

10. A hybridoma comprising a B cell obtained from a transgenic non-human animal having a genome comprising a human heavy chain transgene and a light chain transgene fused to an immortalized cell, wherein the hybridoma produces a detectable amount of the antibody of any one of claims 1 - 5 .

11. A bispecific molecule comprising the antibody of any one of claims 1 - 5 and a second antibody which binds a human antigen presenting cell (APC.)

12. A composition comprising the antibody of any one of claims 1 - 5 and a pharmaceutically acceptable carrier.

13. The composition of claim 12 further comprising a chemotherapeutic agent.

14. An immunotoxin comprising the antibody of any one of claims 1 - 5 linked to a cytotoxic agent.
