Abstract

The present invention relates to optimized Fc variants, methods for their generation, and antibodies and Fc fusions comprising optimized Fc variants.

Claims (15)

1. A protein comprising an Fc variant of a parent Fc polypeptide, wherein said Fc variant comprises an amino acid modification at position 332, wherein numbering is according to the EU index, and wherein said Fc variant comprises an engineered glycoform.

2. A protein comprising an Fc variant of a parent Fc polypeptide, wherein said Fc variant comprises an amino acid modification selected from the group consisting of 332A, 332D, 332E, 332F, 332G, 332H, 332K, 332L, 332M, 332N, 332P, 332Q, 332R, 332S, 332T, 332V, 332W and 332Y, wherein numbering is according to the EU index, and wherein said Fc variant comprises an engineered glycoform.

A protein comprising an Fc variant of a parent Fc polypeptide, wherein said Fc variant comprises an amino acid modification at position 332, wherein said Fc variant further comprises an amino acid substitution at a position selected from the group consisting of 221, 222, 224, 227, 228, 230, 231, 223, 233, 234, 235, 236, 237, 238, 239, 240, 241, 243, 244, 245, 246, 247, 249, 250, 258, 262, 263, 264, 265, 266, 268, 269, 270, 271, 272, 273, 274, 275, 276, 278, 280, 281, 283, 285, 286, 288, 290, 291, 293, 294, 295, 296, 297, 298, 299, 300, 302, 313, 317, 318, 320, 322, 323, 324, 325, 326, 327, 328, 329, 330, 331, 333, 334, 335 336 and 428, wherein numbering is according to the EU index, and wherein said Fc variant comprises an engineered glycoform.

A protein according to claim 1 , wherein said Fc variant further comprises an amino acid modification at a position selected from the group consisting of 235A, 235D, 235E, 235F, 235G, 235H, 235I, 235K, 235M, 235N, 235P, 235Q, 235R, 235S, 235T, 235V, 235W, 235Y, 236A, 236D, 236E, 236F, 236H, 236I, 236K, 236L, 236M, 236N, 236P, 236Q, 236R, 236S, 236T, 236V, 236W, 236Y, 239D, 239E, 239F, 239G, 239H, 239I, 239K, 239L, 239M, 239N, 239P, 239Q, 239R, 239T, 239V, 239W, 239Y, 243E, 243H, 243L, 243Q, 243R, 243W, 243Y, 267D, 267E, 267F, 267H, 267I, 267K, 267L, 267M, 267N, 267P, 267Q, 267R, 267T, 267V, 267W, 267Y, 268D, 268E, 268F, 268G, 268I, 268K, 268L, 268M, 268P, 268Q, 268R, 268T, 268V, 268W, 325A, 325D, 325E, 325F, 325G, 325H, 325I, 325K, 325L, 325M, 325P, 325Q, 325R, 325S, 325T, 325V, 325W, 325Y, 328A, 328D, 328E, 328F, 328G, 328H, 328I, 328K, 328M, 328N, 328P, 328Q, 328R, 328S, 328T, 328V, 328W, 328Y, 330E, 330F, 330G, 330H, 330I, 330L, 330M, 330N, 330P, 330R, 330S, 330T, 330V, 330W, and 330Y, and wherein numbering is according to the EU index.

A protein comprising an Fc variant of a parent Fc polypeptide, wherein said Fc variant comprises an amino acid modification at position 332, wherein said Fc variant has greater affinity for an Fc gamma receptor relative to said parent Fc polypeptide, wherein numbering is according to the EU index, and wherein said Fc variant comprises an engineered glycoform.

6. A protein according to any of claims 1 - 4 or 5 , wherein said protein is an antibody.

7. An antibody according to claim 6 wherein said antibody is selected from the group consisting of a human antibody, a humanized antibody, and a monoclonal antibody.

8. A protein according to any of claims 1 , 2 or 4 further comprising a modification at position 297.

9. A protein according to any of claims 1 - 4 , wherein said engineered glycoform is a reduced level of fucose relative to a native human lgG.

10. A protein according to claim 9 , wherein said protein has been produced in cells wherein a glycosylation enzyme has been modified.

11. A protein according to claim 10 , wherein said protein has been produced in lec13 CHO cells or YB2/0 cells.

12. A protein according to claim 10 , wherein said glycosylation enzyme is α 1,6-fucosyltranserase (FUT8) or β1-4-N-acetylglucosaminyltransferase III (GnTIII).

A protein according to any of claims 1 - 4 wherein said protein has specificity for a target antigen selected from the group consisting of CD19, CD20, CD22, CD30, CD33, CD40, CD40L, CD52, Her2/neu, EGFR, EpCAM, MUC1, GD3, CEA, CA 125, HLA-DR, TNFaipha, MUC18, prostate specific membrane antigen (PMSA) and VEGF.

14. A composition comprising the protein according to any of claims 1 - 4 further comprising a pharmaceuticaily acceptable carrier.

15. A protein according to claim 3 , wherein said protein comprises a modification at position 297.
