Abstract

The invention provides methods for detecting alpha-synuclein. The invention also identifies preferred epitopes of alpha synuclein for use in such detection, and provides antibodies specifically binding to such epitopes.

Claims (27)

1. A method for detecting alpha synuclein in a fluid sample, comprising:

(i) contacting the fluid sample with a capture antibody, whereby the capture antibody captures the alpha synuclein, and

(ii) detecting the alpha synuclein using a reporter antibody,

wherein the capture and reporter antibodies bind to different epitopes on alpha synuclein, and;

wherein the reporter antibody or the capture antibody is an end-specific antibody that binds to the C-terminal end of SN1-119 (residues 1-119 of SEQ ID NO:1) or SN1-122 (residues 1-122 of SEQ ID NO:1).

2. A method for detecting alpha synuclein in a fluid sample, comprising:

(i) contacting the fluid sample with a capture antibody immobilized to a solid phase, and a reporter antibody in solution, whereby the immobilized capture antibody and the reporter antibody bind to the alpha synuclein forming a sandwich, and

(ii) detecting the alpha synuclein using the reporter antibody,

wherein the capture antibody specifically binds to an epitope within amino acids 91-99 of SEQ ID NO:1 and the reporter antibody binds to an epitope within amino acids 109-120 of SEQ ID NO:1.

3. The method of claim 2 , wherein the capture antibody is 1H7 (deposited as ATCC Accession No. PTA-8220) and the reporter antibody is 5C12 (deposited as ATCC Accession No. PTA-9197).

4. The method of claim 3 , wherein the sample is a sample from a human.

5. The method of claim 2 , that is an ELISA method.

6. The method of claim 2 , wherein the capture antibody is immobilized in a microtiter well.

7. A method for detecting alpha synuclein in a fluid sample, comprising:

(i) contacting the fluid sample with a capture antibody immobilized to a support, and a reporter antibody in solution, whereby the immobilized capture antibody and the reporter antibody bind to the alpha synuclein forming a sandwich and

(ii) detecting the alpha synuclein using the reporter antibody,

wherein the capture antibody is 1H7 (deposited as ATCC Accession No. PTA-8220) and the reporter antibody is 9E4 (deposited as ATCC Accession No. PTA-8221).

8. The method of claim 7 , wherein the sample is a sample from a human.

9. A method for detecting alpha synuclein in a fluid sample, comprising:

(i) contacting the fluid sample with a capture antibody immobilized to a support, and a reporter antibody in solution, whereby the immobilized capture antibody and the reporter antibody bind to the alpha synuclein forming a sandwich and

(ii) detecting the alpha synuclein using the reporter antibody,

wherein the capture antibody is 1H7 (deposited as ATCC Accession No. PTA-8220) and the reporter antibody is 11A5 (deposited as ATCC Accession No. PTA-8222).

10. The method of claim 9 , wherein the sample is a sample from a human.

11. A method for detecting alpha synuclein in a fluid sample, comprising:

(i) contacting the fluid sample with a capture antibody immobilized to a support, and a reporter antibody in solution, whereby the immobilized capture antibody and the reporter antibody bind to the alpha synuclein forming a sandwich and

(ii) detecting the alpha synuclein using the reporter antibody, and

wherein the capture and reporter antibodies bind to different epitopes on alpha synuclein, wherein the capture antibody specifically binds to an epitope within SN 91-99 (residues 91-99 of SEQ ID NO:1), and wherein the detecting step is performed twice, wherein the reporter antibody in a first detecting step specifically binds to an epitope within SN 109-120 (residues 109-120 of SEQ ID NO:1), and the reporter antibody in a second detecting step specifically binds to an epitope within SN 120-126 (residues 120-126 of SEQ ID NO:1).
