Abstract

This invention is directed to therapeutic compositions containing non-MHC-restricted T-cells/NK-cells in combination with MHC-restricted T-cells and especially to therapeutic compositions, which comprise LAK cells. Furthermore, the present invention is directed to the use of the above combination in the treatment of tumors in humans, which tumors show a missing, low or aberrant expression of MHC class 1a or 1b molecules. By using the aforementioned compositions/combinations it is possible to provide a balanced selective pressure against emergence of tumor cell variants that would otherwise escape immune detection.

Claims (5)

1. A therapeutic composition comprising:

a) a preparation of activated non-MHC-restricted T cells comprising Lymphokine Activated Killer T cells (LAK T cells), wherein the LAK T cells are prepared by culturing a preparation of peripheral blood mononuclear cells (PBMCs) in a culture medium comprising at least about 1000 units/ml of human interleukin 2 (IL-2) for at least two weeks; and

b) a therapeutic agent for inducing immune responses of MHC-restricted T cells selected from the group consisting of a genetically engineered tumor cell and a vaccine comprising tumor cells or dendritic cells.

2. The therapeutic composition of claim 1 , wherein the culture medium comprises RPMI Medium, 2 mM L-glutamine, 1 mM pyruvate, 100 U/ml penicillin/streptomycin, 15% human serum, and at least about 1000 units/ml of human IL-2.

3. The therapeutic composition of claim 2 , wherein the culture medium further comprises about 1% (vol/vol) of phytohemagglutinin (PHA) during the first 3-4 days of culture.
