Chimeric and humanized antibodies to α5β1 integrin that modulate angiogenesis
The present invention provides chimeric and humanized antibodies that specifically recognize α5β1 integrin, and methods for using the antibodies for reducing or inhibiting angiogenesis in a tissue. Also provided are methods of determining therapeutically acceptable doses of the antibodies and pharmaceutical compositions including the same.
1. An isolated nucleic acid encoding a polypeptide comprising a heavy chain amino acid sequence selected from the group consisting of SEQ ID NOs: 2-6, 16, 20, 25, 28, and 31; and a light chain amino acid sequence selected from the group consisting of SEQ ID NOs: 8-12, 18, 22, 26, and 32.
2. The isolated nucleic acid of claim 1 , wherein the polypeptide comprises the amino acid sequences of SEQ ID NOs:2 and 8.
3. The isolated nucleic acid of claim 1 , wherein the polypeptide comprises the amino acid sequences of SEQ ID NOs:3 and 9.
4. The isolated nucleic acid of claim 1 , wherein the polypeptide comprises the amino acid sequences of SEQ ID NOs:4 and 10.
5. The isolated nucleic acid of claim 1 , wherein the polypeptide comprises the amino acid sequences of SEQ ID NOs:5 and 11.
6. The isolated nucleic acid of claim 1 , wherein the polypeptide comprises the amino acid sequences of SEQ ID NOs:6 and 12.
7. The isolated nucleic acid of claim 1 , wherein the polypeptide comprises the amino acid sequences of SEQ ID NOs:16 and 18.
8. The isolated nucleic acid of claim 1 , wherein the polypeptide comprises the amino acid sequences of SEQ ID NOs:20 and 22.
9. The isolated nucleic acid of claim 1 , wherein the polypeptide comprises the amino acid sequences of SEQ ID NOs:25 and 26.
10. The isolated nucleic acid of claim 1 , wherein the polypeptide comprises the amino acid sequences of SEQ ID NOs:28 and 26.
11. The isolated nucleic acid of claim 1 , wherein the polypeptide comprises the amino acid sequences of SEQ ID NOs:31 and 32.
