Biodegradable cross-linked cationic multi-block copolymers for gene delivery and methods of making thereof
A biodegradable cross-linked cationic multi-block copolymer of linear polyethylenimine (LPEI) wherein the LPEI blocks are linked together by hydrophilic linkers with a biodegradable disulfide bond and methods of making thereof. The biodegradable cross-linked cationic multi-block copolymer may also contain pendant functional moieties which are preferably receptor ligands, membrane permeating agents, endosomolytic agents, nuclear localization sequences, pH sensitive endosomolytic peptides, chromogenic or fluorescent dyes.
1. A process for making a biodegradable water-soluble cross-linked poly(ethyleneamine), the process comprising:
(a) protecting from about 90-95% of the amine nitrogens in a linear poly(ethylenimine) (LPEI) having a molecular weight of from about 1000 Da to about 25000 Da; and
(b) cross-linking the protected LPEI with a cross-linking agent having a linear backbone of from 6-34 backbone members and a reactive group at each end of the backbone, where the backbone includes a disulfide bond.
2. A process according to claim 1 , where the cross-linking is carried out at a molar ratio of cross-linking agent to protected LPEI of from 1:1 to 20:1.
3. A process for making a biodegradable water-soluble cross-linked poly(ethyleneimine), the process comprising:
(a) protecting from about 90-95% of the amine nitrogens in a linear poly(ethylenimine) (LPEI) having a molecular weight of from about 1000 Da to about 25000 Da;
(b) cross-linking the protected LPEI with a cross-linking agent having a linear backbone of from 6-34 backbone members and a reactive group at each end of the backbone, where the backbone includes a disulfide bond; and
(c) reacting the protected cross-linked LPEI with an acylating agent comprising a fatty portion.
4. A process according to claim 3 , where the cross-linking is carried out at a molar ratio of cross-linking agent to protected LPEI of from 1:1 to 20:1.
5. A process according to claim 3 , further comprising (d) reacting the protected cross-linked LPEI with a targeting ligand.
6. A process according to claim 3 where (b) and (c) are carried out in one pot.
7. A process according to claim 3 where the acylating agent comprises a fatty portion coupled to an acylating group via a hydrophilic polyethylene glycol tether.
8. A process according to claim 7 , where the tether has an average molecular weight of less than 1000 Daltons.
9. A process according to claim 1 , further comprising removing the protecting groups.
10. A process according to claim 3 , further comprising removing the protecting groups.
11. A process according to claim 7 where the fatty portion is a oleoyl, palmitoyl, or stearyl group.
12. A process according to claim 7 where the molar ratio of acylating agent to protected LPEI is 1:1 to 40:1.
13. A process according to claim 7 , wherein the cross-linking agent is a dithiodipropionyl halide.
14. A process according to claim 7 , where the cross-linking agent yields a cross-linker group of the formula: —CO(CH 2 ) a SS(CH 2 ) a CO—, where each a is independently an integer from 1 to 15.
15. A process according to claim 7 , where the acylating agent is oleoyltetraethyleneglycolcarbonyl chloride.
