Abstract

The present invention provides compounds of formula (I) wherein R 1 , Y 1 , X 1 , L and R 2 are as defined in the specification, and pharmaceutically acceptable salts thereof, as well as processes for their preparation, pharmaceutical compositions containing them and their use in therapy.

Claims (88)

1. An adenine compound represented by Formula (I):

wherein

R 1 represents hydrogen, hydroxyl, C 1 -C 6 alkoxy, C 2 -C 5 alkoxycarbonyl, or a C 6 -C 10 aryl, C 5 -C 10 heteroaryl or C 3 -C 8 cycloalkyl group, wherein any available carbon atom in each group is optionally substituted by one or more substituents independently selected from halogen, hydroxyl, C 1 -C 6 alkyl, C 1 -C 6 haloalkyl, C 1 -C 6 alkoxy, C 1 -C 6 haloalkoxy, C 2 -C 5 alkoxycarbonyl, amino, mono or di-C 1 -C 6 alkylamino;

Y 1 represents a single bond or C 1 -C 6 alkylene;

X 1 represents a single bond or an oxygen or sulphur atom, sulphinyl, sulphonyl or NR 4 where R 4 is hydrogen or C 1-6 alkyl;

provided that when R 1 is hydroxyl, C 1-6 alkoxy or C 2 -C 5 alkoxycarbonyl and X 1 is oxygen, Y 1 is other than a single bond;

L is a bond or a straight or branched C 1-6 alkylene group, wherein up to 3 carbon atoms within the alkylene group may be replaced by oxygen, sulfur, SO, SO 2 , carbonyl or NR 5 wherein R 5 is hydrogen or C 1-6 alkyl; and

R 2 is either a saturated or partially unsaturated 4-8 membered heterocycle comprising 1 or 2 hetero atoms selected from nitrogen, oxygen and sulphur, provided that at least one of the heteroatoms is nitrogen, which heterocycle is optionally substituted by one or more groups selected from halogen, hydroxyl, oxo, C 1-6 alkoxy, C 2-6 alkylcarbonyl, C 1-6 alkylsulfonyl, carbamoyl, C 1-6 alkylcarbamoyl, di-C 1-6 alkylcarbamoyl, C 1-6 alkylsulfinyl or an C 1-6 alkyl group wherein from 1 to 3 carbon atoms are optionally replaced by oxygen, sulphur, SO, SO 2 , carbonyl or a group NR 3 where R 3 is hydrogen or C 1-6 alkyl;

or R 2 is a group of sub formula (i)

where R 6 is hydrogen or C 1-6 alkyl;

R 7 is hydrogen, or C 1-6 alkyl;

R 8 is hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, a C 6 -C 10 aryl, C 5 -C 10 heteroaryl, C 3 -C 8 cycloalkyl or a 3-8 membered saturated or partially saturated heterocyclic ring wherein each group may be optionally substituted by one or more groups selected from halogen, cyano, S(O) m R 10 , OR 10 , C(O)R 10 , CO 2 R 10 , OC(O)R 10 , SO 2 NR 11 R 12 , CONR 11 R 12 , NR 11 R 12 , NR 11 SO 2 R 10 , NR 11 CO 2 R 10 , NR 11 COR 10 , where any C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl or C 3 -C 8 cycloalkyl groups R 8 may also be optionally substituted by one or more C 6 -C 10 aryl, C 5 -C 10 heteroaryl, a 3-8 membered saturated or partially saturated heterocyclic ring or C 3 -C 8 cycloalkyl groups and where any C 6 -C 10 aryl or C 5 -C 10 heteroaryl groups R 8 may also be optionally substituted by one or more C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl or C 3 -C 8 cycloalkyl groups,

and where any C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 3 -C 8 cycloalkyl group, a C 6 -C 10 aryl, C 5 -C 10 heteroaryl or 3-8 membered saturated or partially saturated heterocyclic ring substituents are optionally substituted by one or more groups independently selected from halogen, cyano, S(O) m R 13 , OR 13 , C(O)R 13 , CO 2 R 13 , OC(O)R 13 , SO 2 NR 14 R 15 , CONR 14 R 15 , NR 14 R 15 , NR 13 SO 2 R 13 , NR 14 CO 2 R 13 or NR 14 COR 13 groups;

or R 7 and R 8 may be combined together with adjacent carbon atom to form a 3-8 membered saturated carbocycle or a 3-8 membered saturated or partially saturated heterocycle optionally substituted by one or more groups independently selected from halogen, cyano, S(O) m R 13 , OR 13 , C(O)R 13 , CO 2 R 13 , OC(O)R 13 , SO 2 NR 14 R 15 , CONR 14 R 15 , NR 14 R 15 , NR 13 SO 2 R 13 , NR 14 CO 2 R 13 or NR 14 COR 13 groups;

R 9 is hydrogen, S(O) m R 16 , C(O)R 16 , SO 2 NR 17 R 18 , CONR 17 R 18 , C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl or C 3 -C 8 cycloalkyl group, the latter four groups being optionally substituted by one or more substituents independently selected from halogen, cyano, S(O) m R 19 , OR 19 , C(O)R 19 , OC(O)R 19 , CO 2 R 19 , SO 2 NR 20 R 21 , CONR 20 R 21 , NR 20 R 21 , NR 20 SO 2 R 19 , NR 20 CO 2 R 19 or NR 20 COR 19 ;

m is 0, 1 or 2;

provided that when R 8 is a C 6 -C 10 aryl or a C 5 -C 10 heteroaryl, R 9 is not hydrogen or unsubstituted C 1 -C 6 alkyl, and

provided that when R 7 and R 9 are hydrogen or C 1 -C 6 alkyl, then R 8 is other than C 1 -C 6 alkyl mono-substituted by C 6 -C 10 aryl or a C 5 -C 10 heteroaryl;

R 10 , R 13 , R 16 and R 19 are independently selected from hydrogen, C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 3 -C 8 cycloalkyl, a C 6 -C 10 aryl or C 5 -C 10 heteroaryl group, which may be optionally substituted on any available carbon atom by one or more groups selected from halogen, hydroxyl, cyano, C 1-6 alkoxy, mercapto, C 1-6 alkylthio, C 1-6 alkylsulphonyl, carboxy, C 1-6 alkylcarbonyloxy, sulphamoyl, C 1-6 alkylsulphamoyl, di-C 1-6 alkylsulphamoyl, carbamoyl, C 1-6 alkylcarbamoyl, di-C 1-6 alkylcarbamoyl, amino, C 1-6 alkylamino or di-C 1-6 alkylamino; and

R 11 , R 12 , R 14 , R 15 , R 17 , R 18 , R 20 or R 21 are independently selected from hydrogen, C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 3 -C 8 cycloalkyl, a C 6 -C 10 aryl or C 5 -C 10 heteroaryl group, may be optionally substituted on any available carbon atom by one or more groups selected from halogen, hydroxyl, cyano, C 1-6 alkoxy, mercapto, C 1-6 alkylthio, C 1-6 alkylsulphonyl, carboxy, C 1-6 alkylcarbonyloxy, sulphamoyl, C 1-6 alkylsulphamoyl, di-C 1-6 alkylsulphamoyl, carbamoyl, C 1-6 alkylcarbamoyl, di-C 1-6 alkylcarbamoyl, amino, C 1-6 alkylamino or di-C 1-6 alkylamino;

or R 11 and R 12 , R 14 and R 15 , R 17 and R 18 or R 20 and R 21 together with the nitrogen atom to which they are attached form a 3- to 8-membered saturated heterocyclic ring comprising a ring nitrogen atom and optionally one or more further heteroatoms independently selected from nitrogen, oxygen, sulphur and sulphonyl, the heterocyclic ring being optionally substituted by one or more substituents independently selected from C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 3 -C 8 cycloalkyl, halogen, hydroxyl, cyano, C 1-6 alkoxy, C 1-6 alkylthio, C 1-6 alkylsulphonyl, sulphamoyl, C 1-6 alkylsulphamoyl, di-C 1-6 alkylsulphamoyl, carbamoyl, C 1-6 alkylcarbamoyl, di-C 1-6 alkylcarbamoyl, amino, C 1-6 alkylamino, di-C 1-6 alkylamino, amido, or a group NR 34 CO 2 R 35 or NR 34 COR 35 where R 34 and R 35 are independently selected from hydrogen and C 1-6 alkyl; or a pharmaceutically acceptable salt thereof.

2. An adenine compound represented by Formula (I):

wherein

R 1 represents hydrogen, hydroxyl, C 1 -C 6 alkoxy, C 2 -C 5 alkoxycarbonyl, or a C 6 -C 10 aryl, C 5 -C 10 heteroaryl or C 3 -C 8 cycloalkyl group, wherein any available carbon atom in each group is optionally substituted by one or more substituents independently selected from halogen, hydroxyl, C 1 -C 6 alkyl, C 1 -C 6 haloalkyl, C 1 -C 6 alkoxy, C 1 -C 6 haloalkoxy, C 2 -C 5 alkoxycarbonyl, amino, mono or di-C 1 -C 6 alkylamino;

Y 1 represents a single bond or C 1 -C 6 alkylene;

X 1 represents a single bond or an oxygen or sulphur atom, sulphinyl, sulphonyl or NR 4 where R 4 is hydrogen or C 1-6 alkyl;

provided that when R 1 is hydroxyl, C 1-6 alkoxy or C 2 -C 5 alkoxycarbonyl and X 1 is oxygen, Y 1 is other than a single bond;

L is a bond or a straight or branched C 1-6 alkylene group, wherein up to 3 carbon atoms within the alkylene group may be replaced by oxygen, sulfur, SO, SO 2 , carbonyl or NR 5 wherein R 5 is hydrogen or C 1-6 alkyl; and

R 2 is either a saturated or partially unsaturated 4-8 membered heterocycle comprising 1 or 2 hetero atoms selected from nitrogen, oxygen and sulphur, provided that at least one of the heteroatoms is nitrogen, which heterocycle is optionally substituted by one or more groups selected from halogen, hydroxyl, oxo, C 1-6 alkoxy, C 2-6 alkylcarbonyl, C 1-6 alkylsulfonyl, carbamoyl, C 1-6 alkylcarbamoyl, di-C 1-6 alkylcarbamoyl, C 1-6 alkylsulfinyl or an C 1-6 alkyl group wherein from 1 to 3 carbon atoms are optionally replaced by oxygen, sulphur, SO, SO 2 , carbonyl or a group NR 3 where R 3 is hydrogen or C 1-6 alkyl;

or R 2 is a group of sub formula (i)

where R 6 is hydrogen or C 1-6 alkyl;

R 7 is hydrogen, or C 1-6 alkyl;

R 8 is hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, a C 6 -C 10 aryl, C 5 -C 10 heteroaryl, C 3 -C 8 cycloalkyl or a 3-8 membered saturated or partially saturated heterocyclic ring wherein each group may be optionally substituted by one or more groups selected from halogen, cyano, S(O) m R 10 , OR 10 , C(O)R 10 , CO 2 R 10 , OC(O)R 10 , SO 2 NR 11 R 12 , CONR 11 R 12 , NR 11 R 12 , NR 11 SO 2 R 10 , NR 11 CO 2 R 10 , NR 11 COR 10 , where any C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl or C 3 -C 8 cycloalkyl groups R 8 may also be optionally substituted by one or more C 6 -C 10 aryl, C 5 -C 10 heteroaryl, a 3-8 membered saturated or partially saturated heterocyclic ring or C 3 -C 8 cycloalkyl groups and where any C 6 -C 10 aryl or C 5 -C 10 heteroaryl groups R 8 may also be optionally substituted by one or more C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl or C 3 -C 8 cycloalkyl groups,

and where any C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 3 -C 8 cycloalkyl group, a C 6 -C 10 aryl, C 5 -C 10 heteroaryl or 3-8 membered saturated or partially saturated heterocyclic ring substituents are optionally substituted by one or more groups independently selected from halogen, cyano, S(O) m R 13 , OR 13 , C(O)R 13 , CO 2 R 13 , OC(O)R 13 , SO 2 NR 14 R 15 , CONR 14 R 15 , NR 14 R 15 , NR 13 SO 2 R 13 , NR 14 CO 2 R 13 or NR 14 COR 13 groups;

or R 7 and R 8 may be combined together with adjacent carbon atom to form a 3-8 membered saturated carbocycle or a 3-8 membered saturated or partially saturated heterocycle optionally substituted by one or more groups independently selected from halogen, cyano, S(O) m R 13 , OR 13 , C(O)R 13 , CO 2 R 13 , OC(O)R 13 , SO 2 NR 14 R 15 , CONR 14 R 15 , NR 14 R 15 , NR 13 SO 2 R 13 , NR 14 CO 2 R 13 or NR 14 COR 13 groups;

R 9 is hydrogen, S(O) m R 16 , C(O)R 16 , SO 2 NR 17 R 18 , CONR 17 R 18 , C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl or C 3 -C 8 cycloalkyl group, the latter four groups being optionally substituted by one or more substituents independently selected from halogen, cyano, S(O) m R 19 , OR 19 , C(O)R 19 , OC(O)R 19 , CO 2 R 19 , SO 2 NR 20 R 21 , CONR 20 R 21 , NR 20 R 21 , NR 20 SO 2 R 19 , NR 20 CO 2 R 19 or NR 20 COR 19 ;

m is 0, 1 or 2;

provided that when R 8 is a C 6 -C 10 aryl or a C 5 -C 10 heteroaryl, R 9 is not hydrogen or unsubstituted C 1 -C 6 alkyl, and

provided that when R 7 and R 9 are hydrogen or C 1 -C 6 alkyl, then R 8 is other than C 1 -C 6 alkyl mono-substituted by C 6 -C 10 aryl or a C 5 -C 10 heteroaryl;

R 10 , R 13 , R 16 and R 19 are independently selected from hydrogen, C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 3 -C 8 cycloalkyl, a C 6 -C 10 aryl or C 5 -C 10 heteroaryl group, which may be optionally substituted on any available carbon atom by one or more groups selected from halogen, hydroxyl, cyano, C 1-6 alkoxy, mercapto, C 1-6 alkylthio, C 1-6 alkylsulphonyl, carboxy, C 1-6 alkylcarbonyloxy, sulphamoyl, C 1-6 alkylsulphamoyl, di-C 1-6 alkylsulphamoyl, carbamoyl, C 1-6 alkylcarbamoyl, di-C 1-6 alkylcarbamoyl, amino, C 1-6 alkylamino or di-C 1-6 alkylamino; and

R 11 , R 12 , R 14 , R 15 , R 17 , R 18 , R 20 or R 21 are independently selected from hydrogen, C- 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 3 -C 8 cycloalkyl, a C 6 -C 10 aryl or C 5 -C 10 heteroaryl group, which may be optionally substituted on any available carbon atom by one or more groups selected from halogen, hydroxyl, cyano, C 1-6 alkoxy, mercapto, C 1-6 alkylthio, C 1-6 alkylsulphonyl, carboxy, C 1-6 alkylcarbonyloxy, sulphamoyl, C 1-6 alkylsulphamoyl, di-C 1-6 alkylsulphamoyl, carbamoyl, C 1-6 alkylcarbamoyl, di-C 1-6 alkylcarbamoyl, amino, C 1-6 alkylamino or di-C 1-6 alkylamino;

or R 11 and R 12 , R 14 and R 15 , R 17 and R 18 or R 20 and R 21 together with the nitrogen atom to which they are attached form a 3- to 8-membered saturated heterocyclic ring comprising a ring nitrogen atom and optionally one or more further heteroatoms independently selected from nitrogen, oxygen, sulphur and sulphonyl, the heterocyclic ring being optionally substituted by one or more substituents independently selected from C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 3 -C 8 cycloalkyl, halogen, hydroxyl, cyano, C 1-6 alkoxy, C 1-6 alkylthio, C 1-6 alkylsulphonyl, sulphamoyl, C 1-6 alkylsulphamoyl, di-C 1-6 alkylsulphamoyl, carbamoyl, C 1-6 alkylcarbamoyl, di-C 1-6 alkylcarbamoyl, amino, C 1-6 alkylamino, di-C 1-6 alkylamino, amido, or a group NR 34 CO 2 R 35 or NR 34 COR 35 where R 34 and R 35 are independently selected from hydrogen and C 1-6 alkyl; provided that when R 1 represents a hydrogen atom, Y 1 represents n-butyl, X 1 represents an oxygen atom and L represents a C 2 alkylene group, then R 2 does not represent a 2-piperidinyl or 4-amino-1-piperidinyl group;

or a pharmaceutically acceptable salt thereof.

3. The compound according to claim 1 wherein Y 1 is unsubstituted C 1-6 alkylene and R 1 is hydrogen, or a pharmaceutically acceptable salt thereof.

4. The compound according to claim 1 wherein X 1 is oxygen, or a pharmaceutically acceptable salt thereof.

5. The compound according to claim 1 wherein L is a C 1-6 alkylene group wherein up to 2 carbon atoms within the alkylene group of L may be replaced by oxygen, sulfur, SO, SO 2 , carbonyl or NR 5 where R 5 is hydrogen or C 1-6 alkyl, or a pharmaceutically acceptable salt thereof.

The compound according to claim 1 or claim 2 wherein R 2 is a saturated or partially unsaturated 4-8 membered heterocycle comprising 1 or 2 hetero atoms selected from nitrogen, oxygen and sulphur, provided that at least one of the heteroatoms is nitrogen, which may be optionally substituted as defined in claim 1 or claim 2 , or a pharmaceutically acceptable salt thereof.

7. The compound according to claim 6 wherein R 2 is optionally substituted pyrrolidine, piperidine, morpholine, thiomorpholine, thiomorpholine-1-oxide or thiomorpholine-1,1-dioxide, or a pharmaceutically acceptable salt thereof.

8. The compound according to claim 1 or claim 2 wherein R 2 is a group of sub formula (i)

where R 6 , R 7 , R 8 and R 9 are as defined in claim 1 or claim 2 , or a pharmaceutically acceptable salt thereof.

9. The compound according to claim 8 wherein R 6 or R 7 are independently hydrogen or C 1-3 alkyl, and R 8 is hydrogen or C 1-6 alkyl optionally substituted as defined in claim 1 or claim 2 , or a pharmaceutically acceptable salt thereof.

The compound according to claim 9 wherein R 8 hydrogen or a C 1-6 alkyl optionally substituted by one or more groups selected from C 2-5 alkoxycarbonyl, carboxy, hydroxy, amino optionally substituted by one or two C 1-6 alkyl groups which may be the same or different, carbamoyl optionally substituted by one or two C 1-6 alkyl groups which may be the same or different, 6-10 membered aryl optionally substituted by halogen, hydroxy, C 1-6 alkyl, C 1-6 alkoxy, C 1-6 haloalkyl, C 1-6 haloalkoxy, or amino optionally substituted by one or two C 1-6 alkyl groups which may be the same or different, or a pharmaceutically acceptable salt thereof.

11. The compound according to claim 8 wherein R 9 is hydrogen or a group CONR 17 R 18 where R 17 and R 18 are as defined in claim 1 or claim 2 , or a pharmaceutically acceptable salt thereof.

12. The compound according to claim 1 which is selected from:

6-Amino-2-butoxy-9-(3-pyrrolidin-1-ylpropyl)-7,9-dihydro-8H-purin-8-one;

1-[3-(6-Amino-2-butoxy-8-oxo-7,8-dihydro-9H-purin-9-yl)propyl]-N,N-dimethyl-L-prolinamide;

6-Amino-2-butoxy-9-{3-[(2S)-2-(methoxymethyl)pyrrolidin-1-yl]propyl}-7,9-dihydro-8H-purin-8-one;

N 2 -[3-(6-Amino-2-butoxy-8-oxo-7,8-dihydro-9H-purin-9-yl)propyl]-N 1 ,N 1 ,N 2 -trimethylglycinamide;

1-[3-(6-Amino-2-butoxy-8-oxo-7,8-dihydro-9H-purin-9-yl)propyl]pyrrolidine-2,5-dione;

6-Amino-2-butoxy-9-{3-[(1S)-2-methoxy-1-methylethyl]amino}propyl)-7,9-dihydro-8H-purin-8-one; and

6-Amino-2-butoxy-9-{3-[methyl-(2-morpholin-4-yl-2-oxo-ethyl)-amino]-propyl}-7,9-dihydro-purin-8-one,

and pharmaceutically acceptable salts thereof.

13. A process for the preparation of a compound of formula (I) or a pharmaceutically acceptable salt thereof as defined in claim 1 which comprises either

(a) reacting a compound of formula (II)

wherein Y 1 , X 1 , R 1 and L are as defined in claim 1 , and Z is a leaving group, with a compound of formula (III)

H—R 2   (III)

where R 2 is as defined in claim 1 , or

(b) reacting a compound of formula (IV)

where L, R 2 , R 1 , X 1 and Y 1 are as defined in claim 1 and R 38 is a C 1-4 alkyl group, with an acid;

or

(c) reacting a compound of formula (VI)

where R 2 and L are as defined in claim 1 and Z 1 is a leaving group, with a compound of formula (VII)

R 1 —Y 1 —X 1 —H  (VII)

where R 1 , Y 1 and X 1 are as defined in claim 1 ; or

(d) reacting a compound of formula (XI)

where R 1 , X 1 , Y 1 , L and R 2 are as defined in claim 1 with a metal alkoxide, followed by treatment with an acid; or

(e) reacting a compound of formula (XIV)

where X 1 , L and R 2 are as defined in claim 1 , with a compound of formula (XV)

where R 1 and Y 1 are as defined in claim 1 and Z 2 is a leaving group; or

(f) reacting a compound of formula (XVII)

where L and R 2 are as defined in claim 1 , with a compound of formula (XVI)

where R 1 , X 1 and Y 1 are as defined in claim 1 ;

and optionally thereafter carrying out one or more of the following:

removal of any protecting groups

forming a pharmaceutically acceptable salt of the compound.

14. A pharmaceutical composition comprising a compound of formula (I) as claimed in claim 1 or a pharmaceutically acceptable salt thereof in association with a pharmaceutically acceptable adjuvant, diluent or carrier.

The compound according to claim 1 or claim 2 wherein R 1 is hydrogen, X 1 is oxygen, L is a straight or branched chain C 1-6 alkylene group, and R 2 is optionally substituted pyrrolidine, piperidine, morpholine, thiomorpholine, thiomorpholine-1-oxide or thiomorpholine-1,1-dioxide, or a pharmaceutically acceptable salt thereof.

16. The compound according to claim 5 wherein L is a straight or branched chain C 1-6 alkylene group, or a pharmaceutically acceptable salt thereof.
