Abstract

The present invention discloses novel compounds of general formula (I) or a pharmaceutically acceptable salt or prodrug thereof (in which X 1 -X 5 , R 5 -R 8b , Z 1 -Z 2 and Ar 1 are defined herein), a method for inhibiting the VR1 receptor in mammals using these compounds, a method for controlling pain in mammals, and pharmaceutical compositions including those compounds and a process for making those compounds.

Claims (78)

1. A compound selected from the group consisting of

N-(5-chloro-2,3-dihydro-1H-inden-1-yl)-N′-1H-indazol-4-ylurea;

N-(5-fluoro-2,3-dihydro-1H-inden-1-yl)-N′-1H-indazol-4-ylurea;

N-(4,5-dimethoxy-2,3-dihydro-1H-inden-1-yl)-N′-1H-indazol-4-ylurea;

N-1H-indazol-4-yl-N′-(5-methoxy-2,3-dihydro-1H-inden-1-yl)urea;

N-(5,6-dimethoxy-2,3-dihydro-1H-inden-1-yl)-N′-1H-indazol-4-ylurea;

N-1H-indazol-4-yl-N′-(6-methoxy-2,3-dihydro-1H-inden-1-yl)urea;

N-[(1S)-4,5-dimethoxy-2,3-dihydro-1H-inden-1-yl]-N′-1H-indazol-4-ylurea;

N-[(1R)-4,5-dimethoxy-2,3-dihydro-1H-inden-1-yl]-N′-1H-indazol-4-ylurea;

N-1H-indazol-4-yl-N′-[(1S)-5-methoxy-2,3-dihydro-1H-inden-1-yl]urea;

N-1H-indazol-4-yl-N′-[(1R)-5-methoxy-2,3-dihydro-1H-inden-1-yl]urea;

N-1H-indazol-4-yl-N′-[5-(2-methoxy-1,1-dimethylethyl)-2,3-dihydro-1H-inden-1-yl]urea;

N-[5-(2-hydroxy-1,1-dimethylethyl)-2,3-dihydro-1H-inden-1-yl]-N′-1H-indazol-4-ylurea;

N-(5-tert-butyl-2-fluoro-2,3-dihydro-1H-inden-1-yl)-N′-1H-indazol-4-ylurea;

N-[(1R,2S)-5-tert-butyl-2-fluoro-2,3-dihydro-1H-inden-1-yl]-N′-1H-indazol-4-ylurea;

N-[(1S,2S)-5-tert-butyl-2-fluoro-2,3-dihydro-1H-inden-1-yl]-N′-1H-indazol-4-ylurea;

N-[(1S,2R)-5-tert-butyl-2-fluoro-2,3-dihydro-1H-inden-1-yl]-N′-1H-indazol-4-ylurea;

N-1H-indazol-4-yl-N′-(6-fluoro-2,3-dihydro-1H-inden-1-yl)urea;

N-1H-indazol-4-yl-N′-(6-methyl-2,3-dihydro-1H-inden-1-yl)urea;

N-(5-tert-butyl-2,2-difluoro-2,3-dihydro-1H-inden-1-yl)-N′-1H-indazol-4-ylurea;

N-1H-indazol-4-yl-N′-[4-(trifluoromethyl)-2,3-dihydro-1H-inden-1-yl]urea;

N-(4-chloro-2,3-dihydro-1H-inden-1-yl)-N′-1H-indazol-4-ylurea;

N-1H-indazol-4-yl-N′-[4-(trifluoromethoxy)-2,3-dihydro-1H-inden-1-yl]urea;

N-(4-bromo-2,3-dihydro-1H-inden-1-yl)-N′-1H-indazol-4-ylurea;

N-[4-(cyanomethyl)-2,3-dihydro-1H-inden-1-yl]-N′-1H-indazol-4-ylurea; and

N-1H-indazol-4-yl-N′-(4-methyl-2,3-dihydro-1H-inden-1-yl)urea;

or a pharmaceutically acceptable salt thereof.

2. A compound that is amorphous N-[(1R)-5-tert-butyl-2,3-dihydro-1H-inden-1-yl]-N′-1H-indazol-4-ylurea, or a pharmaceutically acceptable salt thereof.

3. A compound selected from the group consisting of

N-[(1R)-5-tert-butyl-2,3-dihydro-1H-inden-1-yl]-N′-1H-indazol-4-ylurea, hydrochloride;

N-[(1R)-5-tert-butyl-2,3-dihydro-1H-inden-1-yl]-N′-1H-indazol-4-ylurea, tosylate; and

N-[(1R)-5-tert-butyl-2,3-dihydro-1H-inden-1-yl]-N′-1H-indazol-4-ylurea, benzenesulfonate.

4. A compound selected from the group consisting of

N-(1-acetyl-1H-indazol-4-yl)-N′-(5,6-dimethoxy-2,3-dihydro-1H-inden-1-yl)urea; and

N-(1-acetyl-1H-indazol-4-yl)-N′-[(1R)-5-tert-butyl-2,3-dihydro-1H-inden-1-yl]urea;

or a pharmaceutically acceptable salt thereof.

5. A compound selected from the group consisting of

N-(6-fluoro-5-piperidin-1-yl-2,3-dihydro-1H-inden-1-yl)-N′-1H-indazol-4-ylurea;

N-1H-indazol-4-yl-N′-(4-piperidin-1-yl-2,3-dihydro-1H-inden-1-yl)urea;

N-1H-indazol-4-yl-N′-{5-[4-(trifluoromethyl)piperidin-1-yl]-2,3-dihydro-1H-inden-1-yl}urea;

N-1H-indazol-4-yl-N′-(5-cis-octahydroisoquinolin-2(1H)-yl-2,3-dihydro-1H-inden-1-yl)urea;

N-[5-(1,4′-bipiperidin-1′-yl)-2,3-dihydro-1H-inden-1-yl]-N′-1H-indazol-4-ylurea;

N-1H-indazol-4-yl-N′-[5-(4-phenylpiperidin-1-yl)-2,3-dihydro-1H-inden-1-yl]urea;
