Abstract

A Process for production of 1-(3-(2-(1-benzothiophen-5-yl)ethoxy)propyl)azetidin-3-ol or salts thereof which comprises using as a starting compound as a (phenylthio)acetic acid derivative or salts thereof represented by the general formula: wherein X 1 represents a halogen atom, is useful as a safe process for mass production of 1-(3-(2-(1-benzothiophen-5-yl)ethoxy)propyl)azetidin-3-ol or salts thereof which is useful as a remedy for disease of central and peripheral nerve.

Claims (17)

1. A process for production of 1-(3-(2-(1-benzothiophen-5-yl)ethoxy)propyl)azetidin-3-ol, a salt of 1-(3-(2-(1-benzothiophen-5-yl)ethoxy)propyl)azetidin-3-ol, or a combination thereof, the process comprising:

(A) reducing 1-(3-(2-(1-benzothiophen-5-yl)ethoxy)propionyl)azetidin-3-ol with an activator in the presence of an alkali metal borohydride,

wherein the activator is at least one compound selected from the group consisting of a protonic acid, a methylating agent, and a silylating agent.

2. The process of claim 1 , wherein the activator is sulfuric acid.

3. The process of claim 1 , wherein the alkali metal borohydride is sodium borohydride.

4. The process of claim 2 , wherein an amount of the sulfuric acid employed is 0.5 to 0.6 times mole per mole of the alkali metal borohydride, and

the process comprises adding sulfuric acid at 0 to 30° C. for from 10 minutes to 6 hours, and subsequently reducing at from 30 to 70° C.

5. The process of claim 1 , wherein the activator is a protonic acid.

6. The process of claim 1 , wherein the activator is a methylating agent.

7. The process of claim 1 , wherein the activator is a silylating agent.

8. The process of claim 1 , carried out in a solvent.

9. The process of claim 5 , wherein the activator is at least one protonic acid selected from the group consisting of HCl and H 2 SO 4 .

10. The process of claim 9 , wherein the activator is HCl.

11. The process of claim 5 , wherein the alkali metal borohydride is sodium borohydride.

12. The process of claim 1 , wherein the reducing is in the absence of a boron trifluoride complex.

13. The process of claim 12 , wherein the activator is a protonic acid.

14. The process of claim 12 , wherein the alkali metal borohydride is sodium borohydride.
