Interleukin-2/soluble TGA-beta type II receptor B conjugates and methods and uses thereof
View Patent ↗A conjugate protein comprising an IL-2 and a soluble TGF-beta type II receptor type B and its use in cancer immunotherapy are described.
1. A conjugate protein comprising an interleukin-2 (IL-2) linked to a soluble TGF-beta type II receptor B (sTβRIIB); wherein the sTβRIIB is the ectodomain or extracellular domain of TGF-beta type II receptor B; wherein the conjugate protein comprises the sequence as shown in SEQ ID NO:2, 4, 6, 8 or a variant modified in a linker region between the IL-2 and the sTβRIIB; wherein the variant comprises a sequence having at least 95% similarity to SEQ ID NO:2, 4, 6, 8 and has the same activity as SEQ ID NO:2, 4, 6 or 8.
2. The conjugate protein according to claim 1 , wherein the IL-2 is linked to the sTβRIIB by a peptide linker.
3. A pharmaceutical composition comprising the conjugate protein of claim 1 in admixture with a suitable diluent or carrier.
4. The conjugate protein according to claim 1 , wherein the activity is selected from:
a. dominant negative inhibition of TGF-beta signaling pathway;
b. immunostimulation; and/or
c. angiostatic properties.
