Abstract

The present invention relates to novel Fc variants that comprise at least one novel amino acid residue which may provide for enhanced effector function. More specifically, this invention provides Fc variants that have modified binding affinity to one or more Fc receptor or ligand (e.g., Fc gamma R, C1q). Additionally, the Fc variants have altered complement dependent cytotoxicity (CDC) activity and/or antibody-dependent cell-mediated cytotoxicity (ADCC). The invention further provides methods and protocols for the application of said Fc variants, particularly for therapeutic purposes.

Claims (10)

1. An antibody comprising a variant Fc region of a parent Fc polypeptide, wherein said variant Fc region comprises substitutions at positions 267, 268, and 324, wherein said substitution at position 267 is glutamic acid, said substitution at position 268 is phenylalanine, and said substitution at position 324 is threonine, wherein numbering is according to the EU index as in Kabat.

2. The antibody of claim 1 , wherein said antibody is selected from the group consisting of a human antibody, a humanized antibody, and a monoclonal antibody.

3. The antibody of claim 2 , wherein said antibody is a monoclonal antibody.

4. The antibody of claim 1 , wherein said antibody has specificity for an antigen selected from the group consisting of CD19, CD20, and CD40.

5. The antibody of claim 4 , wherein said antibody has specificity for CD20.

6. An antibody comprising a variant Fc region of a parent Fc polypeptide, wherein said variant Fc region comprises substitutions at positions 267, 268, and 324, wherein said substitution at position 267 is glutamic acid, said substitution at position 268 is phenylalanine, and said substitution at position 324 is threonine, wherein numbering is according to the EU index as in Kabat, and wherein said variant Fc region exhibits greater complement dependent cytotoxicity as compared to said parent Fc polypeptide.

7. The antibody of claim 6 , wherein said antibody is selected from the group consisting of a human antibody, a humanized antibody, and a monoclonal antibody.

8. The antibody of claim 7 , wherein said antibody is a monoclonal antibody.

9. The antibody of claim 6 , wherein said antibody has specificity for an antigen selected from the group consisting of CD19, CD20, and CD40.

10. The antibody of claim 9 , wherein said antibody has specificity for CD20.
