Tumor vaccination in combination with hematopoietic cell transplantation for cancer therapy
View Patent ↗In one aspect, the present invention provides a method for treating cancer comprising tumor cell vaccination in combination with hematopoietic and immune cell transplantation. In some embodiments, the method involves autologous tumor cell vaccination prior to autologous hematopoietic and immune cell transplantation. In another aspect, the present invention provides a method of purifying tumor cells from a subject in preparation for vaccination.
1. A method for treating cancer of a subject comprising:
(a) obtaining purified solid tumor cells from the subject wherein the solid tumor cells are selected from the group consisting of colorectal tumor, lung tumor, breast tumor, pancreatic tumor, liver tumor, prostate tumor, and ovarian tumor;
(b) vaccinating the subject with cancer with said purified solid tumor cells that have been irradiated and formulated in combination with an adjuvant;
(c) collecting immune and hematopoietic cells from the vaccinated subject;
(d) irradiating said subject to myeloablative total body irradiation; and
(e) injecting the collected immune and hematopoietic cells from the subject intravenously into said subject, without ex vivo T cell expansion or treatment with cytokines.
2. The method of claim 1 wherein the tumor cells are purified away from stromal cells.
3. The method of claim 1 wherein the tumor cells are purified away from immunosuppressive cells.
4. The method of claim 1 wherein the purified tumor cells are stimulated with adjuvant prior to the vaccinating.
5. The method of claim 1 , wherein the adjuvant is CpG or GM-CSF.
6. The method of claim 1 wherein the immune and hematopoietic cells collected from the vaccinated subject comprise T cells.
7. The method of claim 1 wherein the immune cells comprise hematopoietic progenitor cells.
8. The method of claim 7 wherein the progenitor cells comprise CD34+ cells.
9. The method of claim 1 wherein the hematopoietic cells are mobilized in the vaccinated subject and enriched prior to the step of injecting the collected immune and hematopoietic cells.
10. The method of claim 1 wherein the recipient receives said total body irradiation with local irradiation of the tumor.
11. The method of claim 1 wherein the cancer is colorectal cancer.
12. The method of claim 1 wherein the tumor cells are from a primary or metastatic tumor.
