Abstract

Compositions containing, and, methods administering, TH302, are useful in treatment of cancer and other hyper-proliferative diseases.

Claims (27)

1. A pharmaceutically acceptable formulation suitable for intravenous administration comprising N,N′-bis(2-bromoethyl)phosphorodiamidic acid (1-methyl-2-nitro-1H-imidazol-5-yl)methyl ester (TH302) or a pharmaceutically acceptable salt thereof, and ethanol.

2. A unit dose of the pharmaceutically acceptable formulation of claim 1 .

3. A method of treating cancer, said method comprising administering a formulation of claim 1 intravenously in an amount in the range of about 8 mg/m 2 to about 800 mg/m 2 to a human patient in need of such treatment.

4. The method of claim 3 , wherein the TH302 is administered at a frequency of at least once per day to once per month.

5. The method of claim 4 , wherein the TH302 is administered at a frequency of at least once per week.

6. The method of claim 3 , wherein the TH302 is administered for a period of at least 1 week.

7. The method of claim 6 , wherein the TH302 is administered for a period of at least 3 weeks.

8. The method of claim 7 , wherein the TH302 is administered for a period of at least 24 weeks.

9. The method of claim 3 , wherein the cancer treated is selected from the group consisting of breast cancer, colorectal cancer, pancreatic cancer, and prostate cancer.

10. The method of claim 3 , wherein the cancer is a metastatic cancer selected from the group consisting of bone marrow cancer, lung cancer, liver cancer, lymph node cancer, and ovarian cancer.

11. The method of claim 3 , wherein the TH302 is administered in combination with another anti cancer agent selected from the group consisting of cisplatin, carboplatin, docetaxel, paclitaxel, gemcitabine, and irinotecan, or is administered in combination with radiation therapy.

12. The method of claim 3 , wherein the patient is also treated with docetaxel.

13. The method of claim 3 , wherein the patient is also treated with irinotecan.

14. The method of claim 3 , wherein the formulation contains not more than 5% of any solvent other than ethanol.

15. The method of claim 3 , wherein TH302 is administered in an amount in the range of about 16 mg/m 2 to about 400 mg/m 2 .

16. The method of claim 3 , wherein TH302 is administered in an amount of at least 240 mg/m 2 .

17. The method of claim 16 , wherein cancer is colorectal cancer.

18. The method of claim 17 , wherein irinotecan is administered in combination with TH302.

19. The method of claim 16 , wherein the cancer is ovarian cancer.

20. The method of claim 16 , wherein the cancer is lung cancer.

21. The method of claim 16 , wherein the cancer is prostate cancer.

22. The method of claim 21 , wherein docetaxel is administered in combination with TH302.

23. The method of claim 16 , wherein the TH302 is administered in combination with radiation therapy.

24. The method of claim 23 , wherein the cancer is lung cancer.

25. The method of claim 16 , wherein the cancer is pancreatic cancer.

26. The method of claim 16 , wherein the cancer is bone or soft tissue cancer.

27. The method of claim 26 , wherein docetaxel is administered in combination with TH302.
