Abstract

A demineralized bone matrix is produced by a process in which a bone body is placed in a first processing solution comprising an acid to demineralize the bone body. The bone body is periodically removed from the first solution at specific time intervals to perform at least one test, such as a compression test, on a mechanical property of the bone body. When the test yields a desired result, the bone body is exposed to a second processing solution that is less acidic than the first, thus minimizing the exposure of the bone body to the harsh acidic conditions of the demineralization phase of the process.

Claims (30)

1. A method for producing a demineralized cancellous bone matrix, the method comprising:

placing a cancellous bone body in a first processing solution comprising an acid at a pH of greater than about pH=0 to demineralize the cancellous bone body;

periodically removing the cancellous bone body from the first processing solution at specific time intervals to perform a compression test on the cancellous bone body, wherein the compression test comprises applying a compressive force in a range from about 10 g-force/cm 2 to about 4000 g-force/cm 2 ; and

after the cancellous bone body is uniformly compressed to less than about 60 percent of its original shape, exposing the cancellous bone body to a second processing solution.

2. The method of claim 1 , wherein the cancellous bone body has a volume greater than about 4 cubic millimeters.

3. The method of claim 1 , wherein the cancellous bone body is fragmented.

4. The method of claim 1 , wherein the second processing solution has a pH greater than about 4.

5. The method of claim 1 , wherein the second processing solution has a pH ranging from about 7 to about 14.

6. The method of claim 1 , wherein the second processing solution comprises a buffer.

7. The method of claim 1 , wherein the second processing solution is aqueous-based.

8. The method of claim 1 , wherein the second processing solution is organic and is capable of neutralizing the acid within and on the cancellous bone body.

9. The method of claim 1 , wherein deterioration, inactivation, or elimination of an intrinsic growth factor within the demineralized cancellous bone matrix is minimized.

10. The method of claim 9 , wherein the intrinsic growth factor is IGF-1.

11. The method of claim 1 , wherein an intrinsic growth factor within the demineralized cancellous bone matrix is maximally preserved.

12. The method of claim 11 , wherein the intrinsic growth factor is IGF-1.

13. The method of claim 1 , wherein deterioration, inactivation, or elimination of intrinsic bone morphogenic proteins within the demineralized cancellous bone matrix is minimized.

14. The method of claim 1 , wherein intrinsic bone morphogenic proteins within the demineralized cancellous bone matrix are maximally preserved.

15. The method of claim 1 , wherein the cancellous bone body is selected from the group consisting of autogenic bone, allogeneic bone, xenogenic bone, and combinations thereof.

16. The method of claim 1 , wherein the demineralized cancellous bone matrix contains less than about two percent of a mineral by weight.

17. The method of claim 16 , wherein the mineral is selected from the group consisting of calcium, magnesium, and phosphate.

18. The method of claim 1 , wherein the cancellous bone body comprises a block.

19. The method of claim 1 , wherein the specific time intervals range from about 5 minutes to about 4 hours.

20. The method of claim 1 , where the cancellous bone body is uniformly compressed to a size ranging from about 5 to about 60 percent of its original shape.

21. The method of claim 1 , wherein the cancellous bone body comprises a strip.

22. The method of claim 21 , wherein a length of the cancellous bone body ranges from about 5 millimeters to about 500 millimeters.

23. The method of claim 1 , wherein the cancellous bone body comprises a block.

24. The method of claim 1 , wherein the compressive force ranges from about 10 g-force/cm 2 to about 1000 g-force/cm 2 .

25. The method of claim 1 , wherein the pH of the first processing solution is from about pH=0 to about pH=0.5.

26. The method of claim 1 , wherein the pH of the first processing solution is about pH=0.2.

27. The method of claim 1 , wherein after exposing the cancellous bone body to the second processing solution, the cancellous bone body contains less than 2 percent residual calcium by weight.
