Abstract

The invention relates to a method for producing ready to use, antigen loaded or unloaded, cryoconserved mature dendritic cells especially for the production of a vaccine containing said dendritic cells, wherein immature dendritic cells are cultivated in the presence of suitable maturation stimuli and the mature dendritic cells thus obtained are frozen. The dendritic cells can be loaded with antigen before freezing. The invention also relates to a vaccine which can be obtained according to the inventive method and to a composition containing frozen, mature dendritic cells which are loaded with antigen.

Claims (10)

1. Mature dendritic cells frozen in freezing medium comprising autologous serum, 5-25% DMSO, and 5-10% glucose.

2. The dendritic cells of claim 1 , wherein said cells are frozen in said freezing medium at a concentration of from 5×10 6 cells/ml to 20×10 6 cells/ml.

3. The dendritic cells of claim 1 , wherein said dendritic cells are antigen-loaded.

4. The dendritic cells of claim 3 , wherein said dendritic cells are loaded with a DNA or RNA molecule which codes for said antigen.

5. The dendritic cells of claim 3 , wherein said dendritic cells are loaded with an RNA molecule that codes for an antigen.

6. The dendritic cells of claim 1 , wherein said cells are frozen in said freezing medium at a concentration of 10×10 6 cells/ml.

7. The dendritic cells of claim 1 , wherein said freezing medium contains from 10% to 20% DMSO.

8. The dendritic cells of claim 1 , wherein said dendritic cells are derived from monocytes.

9. The dendritic cells of claim 1 , wherein said freezing medium contains from 10% to 20% DMSO and 5% glucose.

10. The dendritic cells of claim 1 , wherein said dendritic cells are loaded with an RNA molecule that codes for an antigen and wherein said freezing medium contains 10% DMSO and 5% to 10% glucose.
