Abstract

A composition comprising a main species HER2 antibody that binds to domain II of HER2 and acidic variants thereof is described. Pharmaceutical formulations comprising the composition, and therapeutic uses for the composition are also disclosed.

Claims (19)

1. A composition comprising a main species HER2 antibody that binds to domain II of HER2 and comprises variable light and variable heavy amino acid sequences in SEQ ID Nos. 3 and 4, respectively, and acidic variants thereof comprising disulfide reduced variant and non-reducible variant of the main species antibody.

2. The composition of claim 1 wherein the composition further comprises glycated variant of the main species antibody.

3. The composition of claim 1 further comprising glycated variant of the main species antibody, deamidated variant of the main species antibody, and sialylated variant of the main species antibody.

4. The composition of claim 1 comprising less than about 25% acidic variants.

5. The composition of claim 1 wherein the main species HER2 antibody and the acidic variant are both intact antibodies.

6. The composition of claim 1 wherein the main species HER2 antibody comprises light chain and heavy chain amino acid sequences in SEQ ID Nos. 15 and 16, respectively.

7. The composition of claim 1 further comprising an amino-terminal leader extension variant of the main species antibody.

8. The composition of claim 7 wherein the amino-terminal leader extension comprises VHS-.

9. The composition of claim 8 wherein the amino-terminal leader extension consists of VHS-.

The composition of claim 1 further comprising an amino acid sequence variant of the main species HER2 antibody selected from the group consisting of an antibody comprising a C-terminal lysine residue on one or both heavy chains thereof, and an antibody with one or more oxidized methionine residues.

11. A pharmaceutical formulation comprising the composition of claim 1 in a pharmaceutically acceptable carrier.

12. The pharmaceutical formulation of claim 11 which is sterile.

13. A composition comprising a main species HER2 antibody comprising variable light and variable heavy sequences of SEQ ID Nos. 3 and 4, respectively, and acidic variants of the main species antibody, wherein the acidic variants comprise glycated variant, deamidated variant, disulfide reduced variant, sialylated variant, and non-reducible variant, wherein the acidic variants comprise less than 25% of the composition, and wherein the acidic variants have essentially the same pharmacokinetics as the main species antibody.

14. A pharmaceutical formulation comprising the composition of claim 13 in a pharmaceutically acceptable carrier.

15. A composition comprising a main species HER2 antibody that binds to domain II of HER2 and comprises variable light and variable heavy amino acid sequences in SEQ ID Nos. 3 and 4, respectively, and acidic variant thereof comprising non-reducible variant of the main species antibody.

16. A pharmaceutical composition comprising:

(a) a main species HER2 antibody comprising variable light and variable heavy sequences of SEQ ID Nos. 3 and 4, respectively;

(b) acidic variants of the main species antibody, wherein the acidic variants comprise glycated variant, deamidated variant, disulfide reduced variant, sialylated variant, and non-reducible variant, wherein the acidic variants comprise less than 25% of the composition, and wherein the acidic variants have essentially the same pharmacokinetics as the main species antibody; and

(c) a pharmaceutically acceptable carrier.
