Abstract

This document relates to methods and materials for detecting premalignant and malignant neoplasms. For example, methods and materials for determining whether or not a stool sample from a mammal contains nucleic acid markers or polypeptide markers of a neoplasm are provided.

Claims (14)

1. A method of detecting colorectal cancer or pre-cancer in a mammal, said method comprising:

a) obtaining a stool sample from a mammal; and

b) measuring a K-ras mutation score within said stool sample, measuring BMP3 methylation status within said stool sample, and measuring the level of human DNA within said stool sample,

wherein measurement of an elevated K-ras mutation score, elevated BMP methylation status, and elevated human DNA level as compared to a normal control indicates that said mammal has said colorectal cancer or pre-cancer.

2. The method of claim 1 , wherein said measuring of said K-ras mutation score is measured by digital melt curve analysis.

3. The method of claim 1 , wherein said measuring of said K-ras mutation score is measured by quantitative allele-specific PCR.

4. A method of detecting aero-digestive cancer or pre-cancer in a mammal, said method comprising;

a) obtaining a stool sample from a mammal; and

b) measuring a K-ras mutation score within said stool sample, measuring BMP3 methylation status within said stool sample, and measuring the level of human DNA within said stool sample,

wherein measurement of an elevated K-ras mutation score, elevated BMP methylation status, and elevated human DNA level as compared to a normal control indicates that said mammal has said aero-digestive cancer or pre-cancer.

5. The method of claim 4 , wherein said measuring of said K-ras mutation score is measured by digital melt curve analysis.

6. The method of claim 4 , wherein said measuring of said K-ras mutation score is measured by quantitative allele-specific PCR.

7. The method of claim 4 , wherein said method further comprises determining whether or not a stool sample from said mammal comprises an elevated APC mutation score.

8. The method of claim 7 , wherein said APC mutation score can be measured by digital melt curve analysis.
