Abstract

The present invention relates to spirocarbamate compounds of Formula (I) in which R1, (R2)Y, R3, R4, X and A have the meanings given in the specification. The invention further provides pharmaceutical compositions containing the compounds or pharmaceutically acceptable salts thereof and relates to their use of these compounds as TRPV4 antagonists in treating or preventing conditions associated with TRPV4 imbalance.

Claims (303)

1. A compound of Formula (I):

Wherein:

R 1 is hydrogen, C 1-3 alkyl, CH 2 OH, CH 2 —O—CH 3 , CH 2 OCH 2 Ph, CH 2 CN, CN, halo or C(O)OCH 3 ;

R 2 is independently hydrogen, CN, CF 3 , halo, SO 2 C 1-3 alkyl, C 1-3 alkyl or C≡CH;

R 3 is hydrogen, C 1-2 alkyl, CF 3 or OH;

R 4 is hydrogen, halo or C 1-3 alkyl;

X is CR 4 or N;

A is (CH 2 ) n -Het;

or A is (CH 2 ) n —(CR a R b )—(C 1-2 ) m -Het;

R a is hydrogen or C 1-3 alkyl, wherein the C 1-3 alkyl may be further substituted with one or more halos;

R b is C 1-3 alkyl;

or R a and R b together with the carbon atom they are attached form a C 3-6 cycloalkyl group;

or one of the carbon atoms in the C 3-6 cycloalkyl group formed by R a and R b may be replaced with oxygen to form an oxetane, tetrahydrofuryl or tetrahydropyranyl group;

or one of the carbon atoms in the C 3-6 cycloalkyl group formed by R a and R b may be replaced with nitrogen to form a pyrrolidinyl or piperidinyl group;

Het is:

wherein Het may be substituted by one, two or three substituents chosen from: halo, C 1-5 alkyl, CN, CH 2 F, CHF 2 , CF 3 , C 3-6 cycloalkyl, (CH 2 ) n —O—C 1-3 alkyl, (CH 2 ) n -phenyl, (CH 2 ) n -pyridyl, pyrimidinyl, pyrazinyl, CH(CH 3 )—O—C 1-3 alkyl, CH(OH)—C 1-5 alkyl, C(CH 3 ) 2 —R 5 , C(O)N(CH 3 ) p , N(C 1-3 alkYl) p , NH 2 , C(O)NH 2 , oxetane, oxetane-CH 3 , tetrahydrofuryl, tetrahydropyranyl, morpholinyl, or pyrazolyl;

wherein the phenyl, pyrazolyl, and pyridyl substituent on the Het may be further substituted by one or two substituents chosen from: halo, CN, OCH 3 , C 1-3 alkyl or CF 3 ;

and the C 1-5 alkyl and C 3-6 cycloalkyl substituent on the Het may be further substituted by CN or OH;

R 5 is CN, O—C 1-4 alkyl, (CH 2 ) m —OH, (CH 2 ) p —O—C(O)—O—C 1-5 alkyl, or O—(CH 2 ) p O—R 6 ;

R 6 is C 1-4 alkyl or P(O) 2 (CH 3 ) 2 ;

n is independently 0, 1 or 2;

m is independently 0, 1 or 2;

p is independently 1 or 2; and

y is 1, 2 or 3;

or a pharmaceutically acceptable salt thereof.

2. A compound of Formula (I):

Wherein:

R 1 is hydrogen, C 1-3 alkyl, CH 2 OH, CH 2 —O—CH 3 , CH 2 OCH 2 Ph, CH 2 CN, CN, halo or C(O)OCH 3 ;

R 2 is independently hydrogen, CN, CF 3 , halo, SO 2 C 1-3 alkyl, C 1-3 alkyl or C≡CH;

R 3 is hydrogen, C 1-2 alkyl, CF 3 or OH;

R 4 is hydrogen, halo or C 1-3 alkyl;

X is CR 4 or N;

A is (CH 2 ) n -Het;

or A is (CH 2 ) n —(CR a R b )—(CH 2 ) m -Het;

R a is hydrogen or C 1-3 alkyl, wherein the C 1-3 alkyl may be further substituted with one or more halos;

R b is C 1-3 alkyl;

or R a and R b together with the carbon atom they are attached form a C 3-6 cycloalkyl group;

or one of the carbon atoms in the C 3-6 cycloalkyl group formed by R a and R b may be replaced with oxygen to form an oxetane, tetrahydrofuryl or tetrahydropyranyl group;

or one of the carbon atoms in the C 3-6 cycloalkyl group formed by R a and R b may be replaced with nitrogen to form a pyrrolidinyl or piperidinyl group;

Het is

Wherein Het may be substituted by one, two or three substituents chosen from: halo, C 1-5 alkyl, CN, CH 2 F, CHF 2 , CF 3 , C 3-6 cycloalkyl, (CH 2 ) n —O—alkyl, (CH) n -phenyl, (CH 2 ) n -pyridyl, pyrimidinyl, pyrazinyl, CH(CH 3 )—O—C 1-3 alkyl, C(CH 3 ) 2 —OH, C(CH 3 ) 2 —O—CH 3 , C(CH 3 ) 2 —CN, C(CH 3 ) 2 —CH 2 OH, C(CH 3 ) 2 —CH 2 —O—C(O)—O—C 1-5 alkyl, C(O)N(CH 3 ) p , N(C 1-3 alkyl) p , NH 2 , C(O)NH 2 , oxetane, oxetane-CH 3 , tetrahydrofuryl or tetrahydropyranyl;

wherein the phenyl and pyridyl substituent on the Het may be further substituted by one or two substituents chosen from: halo, CN, OCH 3 , C 1-3 alkyl or CF 3 ;

and the C 1-6 alkyl and C 3-6 cycloalkyl substituent on the Het may be further substituted by CN or OH;

n is independently 0, 1 or 2;

m is independently 0, 1 or 2;

p is independently 1 or 2;

y is 1, 2 or 3;

or a pharmaceutically acceptable salt thereof.

3. A compound of claim 2 wherein:

R 1 is hydrogen, C 1-3 alkyl or CH 2 OH;

R 2 is CN;

R 3 is hydrogen;

X is N;

A is (CH 2 ) n -Het;

or A is (CH 2 ) n —(CR a R b )—(CH 2 ) m -Het;

R a is hydrogen or C 1-3 alkyl, wherein the C 1-3 alkyl may be further substituted with one or more halos;

R b is C 1-3 alkyl;

or R a and R b together with the carbon atom they are attached to form a C 3-6 cycloalkyl group;

or one of the carbon atoms in the C 3-6 cycloalkyl group formed by R a and R b may be replaced with oxygen to form an oxetane, tetrahydrofuryl or tetrahydropyranyl group;

or one of the carbon atoms in the C 3-6 cycloalkyl group formed by R a and R b may be replaced with nitrogen to form a pyrrolidinyl or piperidinyl group;

and the C 1-5 alkyl and C 3-6 cycloalkyl substituent on the Het may be further substituted by CN or OH;

n is independently 0 or 1;

m is independently 0 or 1;

p is independently 1 or 2; and

y is 1 or 2;

4. A compound of claim 2 wherein:

R 1 is hydrogen, C 1-3 alkyl or CH 2 OH;

R 2 is CN;

R 3 is hydrogen;

X is N;

A is (CH 2 ) n -Het;

or A is (CH 2 ) n —(CR a R b )—(CH 2 ) m -Het;

R a is hydrogen or C 1-3 alkyl, wherein the C 1-3 alkyl may be further substituted with one or more halos;

R b is C 1-3 alkyl;

or R a and R b together with the carbon atom they are attached to form a C 3-6 cycloalkyl group;

or one of the carbon atoms in the C 3-6 cycloalkyl group formed by R a and R b may be replaced with oxygen to form an oxetane, tetrahydrofuryl or tetrahydropyranyl group;

or one of the carbon atoms in the C 3-6 cycloalkyl group formed by R a and R b may be replaced with nitrogen to form a pyrrolidinyl or piperidinyl group;

5. A compound of claim 2 wherein:

R 1 is hydrogen, C 1-3 alkyl or CH 2 OH;

R 2 is CN;

R 3 is hydrogen;

X is N;

A is (CH 2 ) n -Het;

Het is

wherein Het may be substituted by one, two or three substituents chosen from: halo, C 1-5 alkyl, CN, CH 2 F, CHF 2 , CF 3 , C 3-6 cycloalkyl, (CH 2 ) n —O—C 1-3 alkyl, (CH 2 ) n -phenyl, (CH 2 ) n -pyridyl, pyrimidinyl, pyrazinyl, CH(CH 3 )—O—C 1-3 alkyl, C(CH 3 ) 2 —OH, C(CH 3 ) 2 —O—CH 3 , C(CH 3 ) 2 —CN, C(CH 3 ) 2 —CH 2 OH, C(CH 3 ) 2 —CH 2 —O—C(O)—O—C 1 - 5 alkyl, C(O)N(CH 3 ) p , N(C 1-3 alkyl) p , NH 2 , C(O)NH 2 , oxetane, oxetane-CH 3 , tetrahydrofuryl or tetrahydropyranyl;

and the C 1-5 alkyl and C 3-6 cycloalkyl substituent on the Het may be further substituted by CN or OH;

n is independently 0 or 1;

p is independently 1 or 2; and

y is 1 or 2;

or a pharmaceutically acceptable salt thereof.

6. A compound of claim 2 wherein:

R 1 is hydrogen, C 1-3 alkyl or CH 2 OH;

R 2 is CN;

R 3 is hydrogen;

X is N;

A is (CH 2 ) n -Het;

Wherein Het may be substituted by one, two or three substituents chosen from: Halo, C 1-5 alkyl, CN, CH 2 F, CHF 2 , CF 3 , C 3-6 cycloalkyl, (CH 2 ) n —O—C 1-3 alkyl, (CH 2 ) n -phenyl, (CH 2 ) n -pyridyl, pyrimidinyl, pyrazinyl, CH(CH 3 )—O—C 1-3 alkyl, C(CH 3 ) 2 —OH, C(CH 3 ) 2 —O—CH 3 , C(CH 3 ) 2 —CN, C(CH 3 ) 2 —CH 2 OH, C(CH 3 ) 2 —CH 2 —O—C(O)—O—C 1 - 5 alkyl, C(O)N(CH 3 ) p , N(C 1 - 3 alkyl) p , N H 2 , C(O)NH 2 , oxetane, oxetane-CH 3 , tetrahydrofuryl or tetrahydropyranyl;

1-(((5S,7S)-3-(3-(2-cyanopropan-2-yl)isoxazol-5-yl)-7-methyl-2-oxo-1-oxa-3-azaspiro[4.5]decan-7-yl)methyl)-1H-benzo[d]imidazole-6-carbonitrile;

8. A compound which is 1-(((5S,7S)-3-(5-Ethoxypyrazin-2-yl)-7-methyl-2-oxo-1-oxa-3-azaspiro[4.5]decan-7-yl)methyl)-1H-benzo[d]imidazole-6-carbonitrile;

or a pharmaceutically acceptable salt thereof.

9. A compound which is 1-(((5S,7S)-3-(5-(2-hydroxypropan-2-yl)pyrazin-2-yl)-7-methyl-2-oxo-1-oxa-3-azaspiro[4.5]decan-7-yl)methyl)-1H-benzo[d]imidazole-6-carbonitrile;

or a pharmaceutically acceptable salt thereof.

10. A pharmaceutical composition comprising a compound of claim 1 and a pharmaceutically acceptable carrier or excipient.

A method of treating atherosclerosis, disorders related to intestinal edema, post-surgical abdominal edema, local and systemic edema, fluid retention, sepsis, hypertension, inflammation, bone related dysfunctions and congestive heart failure, pulmonary disorders, chronic obstructive pulmonary disorder, ventilator induced lung injury, high altitude induced pulmonary edema, acute respiratory distress syndrome, acute lung injury, pulmonary fibrosis, sinusitis/rhinitis, asthma, pain, motor neuron disorders, genetic gain of function disorders, renal dysfunction, osteoarthritis crohn's disease, colitis, intestinal irregularity (hyperreactivity/hyporeactivity), intestinal pain and cramping, which comprises administering to a human in need thereof, a compound of claim 1 .

12. A method according to claim 11 wherein the compound is administered orally.

13. A method according to claim 11 wherein the compound is administered intravenously.

14. A method according to claim 11 wherein the disease is congestive heart failure.

15. A method according to claim 11 wherein the disease is acute lung injury.

16. A pharmaceutical composition comprising a compound of claim 8 and a pharmaceutically acceptable carrier or excipient.

17. A pharmaceutical composition comprising a compound of claim 9 and a pharmaceutically acceptable carrier or excipient.

20. A method according to claim 15 wherein the compound is administered by inhalation.
