Fc variants with altered binding to FcRn
The present application relates to optimized IgG immunoglobulin engineering methods for their generation, and their application, particularly for therapeutic purposes.
1. A polypeptide comprising a variant Fc region as compared to a parent Fc polypeptide, said variant Fc region comprising an amino acid substitution at position 308, wherein said amino acid substitution is selected from the group consisting of a phenylalanine, a cysteine, a tyrosine and a tryptophan, wherein said polypeptide has increased in vivo half-life as compared to a polypeptide comprising said parent Fc polypeptide, wherein said polypeptide is selected from the group consisting of an antibody and an immunoadhesin and wherein numbering is according to the EU Index in Kabat et al.
2. A polypeptide according to claim 1 which is an antibody.
3. An antibody according to claim 2 wherein said antibody is selected from the group consisting of a chimeric antibody, a humanized antibody, or a human antibody.
4. An antibody according to claim 2 wherein said antibody has specificity for an antigen selected from the group consisting of vascular endothelial growth factor (VEGF), tumor necrosis factor-α (TNF-α), CD25, nerve growth factor (NGF), IgE and epidermal growth factor receptor (EGFR).
5. A composition comprising the polypeptide of claim 1 and a carrier.
