Abstract

Automated systems and methods for providing platelet concentrates and synthetic storage media with reduced residual plasma volumes are disclosed. The disclosed systems and methods reduce the residual volume of plasma in platelet concentrate to obtain a platelet product having a volume of plasma that is approximately 5% or less of the total platelet product volume. The disclosed systems and methods also reduce the residual volume of plasma in platelet concentrate to obtain a washed platelet product, wherein the volume of plasma in the washed platelet product is approximately 1% or less of the total washed platelet product volume. Storage media for platelets including less than approximately 10% plasma are also disclosed.

Claims (25)

1. A method for providing a substantially plasma-reduced platelet product comprising:

introducing a biological fluid including platelets into a separation chamber of an automated apheresis separator;

separating platelets and plasma from the remainder of said biological fluid to obtain platelets with residual plasma;

removing said platelets with residual plasma from said separation chamber and simultaneously introducing said platelets with residual plasma into a concentration chamber different from said separation chamber;

concentrating platelets in said concentration chamber to obtain a platelet concentrate comprising platelets suspended in a reduced volume of residual plasma;

removing a selected volume of plasma from said concentration chamber;

reconstituting the platelet concentrate with an aqueous additive solution to obtain a platelet product comprising platelets, aqueous additive solution and plasma, with the volume of plasma in said platelet product is 5% or less of the total platelet product volume.

2. The method of claim 1 wherein said platelet concentrate is reconstituted outside of said concentration chamber.

3. The method of claim 1 comprising collecting said platelets in said platelet concentration chamber.

4. The method of claim 1 further comprising:

introducing said platelet product into an automated apheresis separator,

removing additional plasma and aqueous additive solution from said platelet product to obtain a second platelet concentrate suspended in a residual volume of plasma and aqueous additive solution,

reconstituting said platelet concentrate in an aqueous additive solution to obtain a platelet product comprising platelets, aqueous additive solution and plasma, wherein the volume of plasma in said product is 1% or less of the total platelet product.

5. The method of claim 4 comprising introducing said platelet product into the same apheresis separator in which the platelet product was obtained.

6. The method of claim 4 further comprising:

introducing a wash solution into said separator,

removing said residual plasma and wash solution from said platelet product to obtain a washed platelet concentrate prior to reconstituting said platelet concentrate.

7. The method of claim 6 wherein said wash solution comprises one of either saline or an aqueous synthetic platelet storage media.

8. The method of claim 6 wherein said wash solution comprises sodium citrate, sodium acetate, sodium phosphate, sodium chloride and glucose.

9. The method of claim 1 wherein said additive solution comprises one of either saline or an aqueous synthetic platelet storage media.

10. The method of claim 1 further comprising removing an additional selected volume of plasma from said concentration chamber.

11. The method of claim 10 wherein said additional selected volume of plasma is approximately 10 ml.

12. The method of claim 1 wherein said aqueous additive solution comprises sodium citrate, sodium acetate, sodium phosphate, sodium chloride and glucose.

13. The method of claim 12 wherein said aqueous additive solution further comprises sodium bicarbonate.

14. The method of claim 1 wherein said biological fluid including platelets comprises platelet-rich plasma.
