Methods for using extracellular adenosine inhibitors and adenosine receptor inhibitors to enhance immune response and inflammation
View Patent ↗A method is provided herein to increase an immune response to an antigen. The method includes administering an agent that inhibits extracellular adenosine or inhibits adenosine receptors. Also disclosed are methods to increase the efficacy of a vaccine and to increase an immune response to a tumor antigen or immune cell-mediated tumor destruction.
1. A method for reducing tumor volume and/or tumor size in a subject, comprising:
administering to the subject
(a) a therapeutically effective dose of an adenosine A2a receptor antagonist, or a therapeutically effective dose of an agent at inhibits accumulation of extracellular adenosine, or combinations thereof, and
(b) a therapeutically effective dose of an anti-neoplastic agent, thereby reducing the volume and/or size of the tumor.
2. The method of claim 1 , wherein the subject is administered the anti-neoplastic agent prior to the adenosine A2a receptor antagonist.
3. The method of claim 1 , wherein the anti-neoplastic agent selectively targets the tumor cells.

The method of claim 1 , wherein the anti-neoplastic agent comprises an alkylating drug, a folate antagonist, a purine antagonist, a pyrimidine antagonist, a spindle poison, a podophyllotoxin, an antibiotic, a nitrosurea, an inorganic ion, a biologic response modifier, an enzyme, a hormone, an immunotoxin that selectively binds to a tumor-specific antigen and accumulates in a tumor, an agent that has an affinity for tumor cells, or an antibody that binds a tumor-specific antigen and has antitumor activity in vivo.
5. The method of claim 1 , wherein the anti-neoplastic agent comprises cyclophosphamide, ifosamide, cisplatin, methotrexate, cytoxan, procarizine, etoposide, BCNU, vincristine, vinblastine, cyclophosphamide, gencitabine, 5-flurouracie, paclitaxel, or doxorubicin.
6. The method of claim 1 , further comprising administering to the subject a therapeutically effective dose of an immunostimulant.
7. The method of claim 1 , further comprising administering to the subject a therapeutically effective dose of radiation therapy.
8. The method of claim 1 , further comprising administering to the subject a therapeutically effective dose of an immunostimulant and an immunotherapy agent that initiates an immune response directed against the tumor.
9. The method of claim 8 , wherein the immunotherapy agent initiates an immune response directed against the tumor by depleting immunoregulatory mechanisms that improve anti-tumor CTL activity.
10. The method of claim 9 , wherein the immunotherapy agent is a CTLA4 ligand or is an agent that removes CD25+ regulatory T cells.
11. The method of claim 1 , further comprising administering to the subject a therapeutically effective dose of a monoclonal antibody that binds to a tumor-specific antigen and initiates anti-tumor activity in vivo.
12. The method of claim 11 , wherein the monoclonal antibody is an immunotoxin.
13. The method of claim 1 , wherein the adenosine A2a receptor antagonist is ZM241385(4(2-[7- amino-2-(2-furyl)[1,2,4]triazolo[ 2 , 3 -α][1,3,5]triazin-5yl-amino]ethyl)phenol), 1,3,7-trimethylxanthine (caffeine), theobromine, SCH 58261, or KW-6002.
14. The method of claim 1 , wherein the agent that inhibits accumulation of extracellular adenosine comprises an inhibitor of hypoxia-induced enzyme.
15. The method of claim 1 , wherein the agent that inhibits accumulation of extracellular adenosine comprises an oxygenation agent, a redox-potential changing agent, adenosine deaminase, adenosine kinase, ADA-PEG, oxygenation of the subject, or combinations thereof.
16. The method of claim 1 , wherein the subject previously received chemotherapy or radiotherapy.
17. The method of claim 1 , wherein the subject is a human.
18. The method of claim 1 , wherein the subject is immunosuppressed.
19. The method of claim 1 , wherein the method:
reduces a number of tumor cells in the subject;
reduces metastasis of the tumor in the subject;
reduces a blood supply of a tumor in a subject; or
combinations thereof.
20. The method of claim 1 , wherein the tumor is greater than 2 mm in diameter, and wherein the tumor has local hypoxia areas.
21. The method of claim 1 , wherein the tumor is a cancer.
22. The method of claim 1 , wherein the cancer is a melanoma, sarcoma, or carcinoma of the colon, breast, cervix, endometrium, ovary, testis, nasopharynx, kidney, lung, skin, or liver.
23. The method of claim 21 , wherein the cancer is a cancer of the lung, breast, skin, or liver.
24. The method of claim 17 , wherein the tumor is a cancer of the lung.
25. The method of claim 17 , wherein the tumor is a cancer of the breast.
