Abstract

The invention provides methods for the synthesis of oligosaccharides comprising an aminooxy group. The invention further provides oligosaccharides comprising an aminooxy group, methods for coupling oligosaccharides comprising an aminooxy group to glycoproteins, and oligosaccharide-protein conjugates. Also provided are methods of treating a lysosomal storage disorder in a mammal by administration of an oligosaccharide-protein conjugate.

Claims (29)

1. A method of preparing an oligosaccharide comprising an aminooxy group, the method comprising:

(a) providing an oligosaccharide, wherein the oligosaccharide is

(b) providing an aminooxy compound, wherein the aminooxy compound is Formula III:

wherein n is chosen from integers ranging from 1 to 10,

P is chosen from amino protecting groups, and

Y is a group selected from carboxyl, activated ester, acyl halide, acyl azide, alkyl halide, anhydride, isothiocyanate, isocyanate, and sulfonyl halide; and

(c) reacting the oligosaccharide with the aminooxy compound,

thereby preparing the oligosaccharide comprising an aminooxy group.

2. The method of claim 1 , wherein Y is

and wherein X is selected from hydroxy, aryloxy, heteroaryloxy, and heterocyclyloxy.

5. The method of claim 4 , wherein step (c) comprises reacting the oligosaccharide with the aminooxy compound in the presence of a coupling reagent and/or a catalyst.

7. A method of preparing an oligosaccharide comprising an aminooxy group, the method comprising:

(a) providing an oligosaccharide, wherein the oligosaccharide is

(b) providing an aminooxy compound, wherein the aminooxy compound is of Formula III:

wherein n is chosen from integers ranging from 1 to 10,

P is an amino protecting groups, and

Y is selected from hydrazine, hydrazide, aminooxy, thiosemicarbazide, semicarbazide, and amino group; and

(c) reacting the oligosaccharide with the aminooxy compound,

thereby preparing the oligosaccharide comprising an aminooxy group.

8. The method of claim 7 , wherein the aminooxy compound is

9. The method of claim 8 , wherein step (c) comprises reacting the oligosaccharide with the aminooxy compound in the presence of a coupling reagent and/or a catalyst.

10. The method of claim 9 , wherein the coupling reagent is 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide (EDC) and the catalyst is N-hydroxysuccinimide (NHS).

11. The method of claim 1 , further comprising

(d) deprotecting the oligosaccharide comprising an aminooxy group.

12. The method of claim 7 , further comprising

(d) deprotecting the oligosaccharide comprising an aminooxy group.
