Abstract

The invention relates to antibodies, and antigen binding fragments thereof, that neutralize infection of both RSV, MPV and PVM. The invention also relates to nucleic acids that encode, immortalized B cells and cultured plasma cells that produce, and to polypeptides that bind to such antibodies and antibody fragments. In addition, the invention relates to the use of the antibodies, antibody fragments, and polypeptides recognized by the antibodies of the invention in screening methods as well as in the diagnosis, treatment and prevention of RSV or MPV infection and RSV and MPV co-infection.

Claims (26)

1. An isolated, purified or recombinant antibody, or an antigen binding fragment thereof comprising the heavy chain CDR1, CDR2, and CDR3 sequences as set forth in SEQ ID NO: 1, SEQ ID NO: 2, and SEQ ID NO: 3, respectively, and the light chain CDR1, CDR2, and CDR3 sequences as set forth in SEQ ID NO: 4, SEQ ID NO: 5, and SEQ ID NO: 35, respectively, wherein the antibody or antigen binding fragment neutralize infection of RSV, MPV and PVM.

2. The antibody of claim 1 , or an antigen binding fragment thereof, that specifically binds a conserved region on the amino terminal portion of the F protein of RSV, MPV, or PVM comprising the amino acid sequence as set forth in any one of SEQ ID NOs: 64-69.

3. The antibody of claim 1 , or an antigen binding fragment thereof, wherein the antibody or antigen binding fragment neutralize infection of both group A and group B RSV as well as both group A and group B MPV.

4. The antibody of claim 1 , or an antigen binding fragment thereof, wherein the antibody or antigen binding fragment neutralize infection of MPV subgroups A1, A2, B1 and B2.

5. The antibody of claim 1 , or an antigen binding fragment thereof, wherein the antibody or antigen binding fragment do not bind to the F protein of RSV at a site that overlaps antigenic site I, antigenic site II, or antigenic site IV.

6. The antibody of claim 1 , or an antigen binding fragment thereof, wherein the concentration of antibody or fragment required for 50% neutralization of RSV, MPV or PVM is 500 ng/ml or less.

The antibody of claim 1 , or an antigen binding fragment thereof, wherein the antibody comprises: a heavy chain variable region having at least 80% sequence identity to the amino acid sequence of SEQ ID NO: 17 and a light chain variable region having at least 80% sequence identity to the amino acid sequence of SEQ ID NO: 37.

An isolated, purified or recombinant antibody, or an antigen binding fragment thereof, wherein the antibody comprises a heavy chain variable region comprising the amino acid sequence of SEQ ID NO: 17 and a light chain variable region comprising the amino acid sequence of SEQ ID NO: 37, wherein the antibody or antigen binding fragment neutralize infection of RSV, MPV and PVM.

9. The antibody of claim 1 , or an antigen binding fragment thereof, wherein the antibody is HMB3210 variant 3.

10. The antibody of claim 1 , or an antigen binding fragment thereof, wherein the antibody is a human antibody, a monoclonal antibody, a human monoclonal antibody, a purified antibody, a single chain antibody, Fab, Fab′, F(ab′)2, Fv or scFv.

11. A method of treating or attenuating infection by RSV or MPV or both RSV and MPV, comprising: administering to a subject in need thereof, a therapeutically effective amount of the antibody of claim 1 , or an antigen binding fragment thereof.

12. A nucleic acid molecule comprising a polynucleotide encoding the antibody, or an antigen binding fragment thereof, of the antibody of claim 1 .

13. The nucleic acid molecule of claim 12 , wherein the polynucleotide sequence is at least 75% identical to the nucleic acid sequence of any one of SEQ ID NOs: 7-11, 16, 36 or 38.

14. A vector comprising the nucleic acid molecule of claim 12 .

15. A cell expressing the antibody of claim 1 , or an antigen binding fragment thereof.

A pharmaceutical composition comprising the antibody of claim 1 , or an antigen binding fragment thereof; a nucleic acid molecule comprising a polynucleotide encoding said antibody, or an antigen binding fragment thereof; a vector comprising said polynucleotide; a cell expressing said vector and a pharmaceutically acceptable diluent or carrier.

A pharmaceutical composition comprising a first antibody or an antigen binding fragment thereof, and a second antibody, or an antigen binding fragment thereof, wherein the first antibody is the antibody of claim 1 , and the second antibody neutralizes infection of RSV or MPV or both RSV and MPV.

18. A method of reducing RSV or MPV infection, or lowering the risk of RSV or MPV infection, comprising: administering to a subject in need thereof, a therapeutically effective amount of the antibody of claim 1 , or an antigen binding fragment thereof.

19. A method of treating or attenuating infection by RSV or MPV or both RSV and MPV, comprising: administering to a subject in need thereof, a therapeutically effective amount of the antibody of claim 2 , or an antigen binding fragment thereof.

20. A nucleic acid molecule comprising a polynucleotide encoding the antibody, or an antigen binding fragment thereof, of the antibody of claim 2 .

21. A pharmaceutical composition comprising the antibody of claim 2 , or an antigen binding fragment thereof, and a pharmaceutically acceptable diluent or carrier.

A method of treating or attenuating infection by RSV or MPV or both RSV and MPV, comprising: administering to a subject in need thereof, a therapeutically effective amount of an antibody, or an antigen binding fragment thereof comprising the heavy chain CDR1, CDR2, and CDR3 sequences as set forth in SEQ ID NO: 1, SEQ ID NO: 2, and SEQ ID NO: 3, respectively, and the light chain CDR1, CDR2, and CDR3 sequences as set forth in SEQ ID NO: 4, SEQ ID NO: 5, and SEQ ID NO: 35, respectively, wherein the antibody or antigen binding fragment neutralize infection of RSV, MPV and PVM.

A pharmaceutical composition comprising an antibody, or an antigen binding fragment thereof comprising (i) the heavy chain CDR1, CDR2, and CDR3 sequences as set forth in SEQ ID NO: 1, SEQ ID NO: 2, and SEQ ID NO: 3, respectively, and the light chain CDR1, CDR2, and CDR3 sequences as set forth in SEQ ID NO: 4, SEQ ID NO: 5, and SEQ ID NO: 35, respectively, wherein the antibody or antigen binding fragment neutralize infection of RSV, MPV and PVM, and a pharmaceutically acceptable diluent or carrier.

A method of treating or attenuating infection by RSV or MPV or both RSV and MPV, comprising: administering to a subject in need thereof, a therapeutically effective amount of an antibody, or an antigen binding fragment thereof, wherein the antibody comprises a heavy chain variable region comprising the amino acid sequence of SEQ ID NO: 17 and a light chain variable region comprising the amino acid sequence of SEQ ID NO: 37, wherein the antibody or antigen binding fragment neutralize infection of RSV, MPV and PVM.

25. A nucleic acid molecule comprising a polynucleotide encoding the antibody, or an antigen binding fragment thereof, of the antibody of claim 8 .

A pharmaceutical composition comprising an antibody, or an antigen binding fragment thereof, wherein the antibody comprises a heavy chain variable region comprising the amino acid sequence of SEQ ID NO: 17 and a light chain variable region comprising the amino acid sequence of SEQ ID NO: 37, wherein the antibody or antigen binding fragment neutralize infection of RSV, MPV and PVM, and a pharmaceutically acceptable diluent or carrier.
