Abstract

The disclosure pertains to antibodies and binding fragments thereof that specifically binds all or part of EHAEVVFTA. Also provided are isolated peptides, isolated nucleic acids, immunogens, compositions, immunoassays and kits and method of using said reagents to detect misfolded TTR.

Claims (26)

1. A conformation specific antibody or binding fragment thereof that specifically binds all of EHAEVVFTA (SEQ ID NO: 1), wherein the antibody or binding fragment thereof is capable of binding to TTR amyloid fibrils as found in situ in a biological sample obtained from a subject.

2. The antibody or binding fragment thereof of claim 1 , wherein the antibody or binding fragment selectively binds TTR fibrils at least 2 fold more efficiently than it binds natively folded tetramer TTR.

3. The antibody or binding fragment thereof of claim 1 , wherein the antibody or binding fragment thereof is capable of disrupting or reducing TTR fibril formation when misfolded TTR intermediates in solution are contacted with the antibody or binding fragment.

4. The antibody or binding fragment thereof of claim 1 , wherein the antibody is a monoclonal or polyclonal antibody and wherein the antibody is further optionally chimeric or humanized.

5. The antibody or binding fragment thereof of claim 1 , wherein the antibody or binding fragment thereof is a monoclonal antibody or a binding fragment thereof.

6. The antibody binding fragment of claim 1 , wherein the antibody fragment is a Fab, Fab′, F(ab′)2, scFv, dsFv, ds-scFv, dimer, minibody, diabody, or multimer thereof or a bispecific antibody fragment.

7. A composition comprising the antibody of claim 1 and a suitable carrier.

8. A kit comprising:

a) the antibody of claim 1 ,

b) a compartment for housing the antibody and

c) optionally instructions.

9. The kit of claim 8 , wherein the compartment for housing the antibody is a vial.

10. A conformation specific antibody or binding fragment thereof that specifically binds all of EHAEVVFTA (SEQ ID NO: 1), wherein the antibody or binding fragment thereof selectively binds a misfolded TTR conformation at least 2 fold more efficiently than it binds natively folded tetramer TTR.

11. The antibody or binding fragment of claim 10 , wherein the antibody or binding fragment selectively binds TTR fibrils.

12. The antibody or binding fragment of claim 10 , wherein the antibody or binding fragment thereof is capable of disrupting or reducing TTR fibril formation when misfolded TTR intermediates in solution are contacted with the antibody or binding fragment.

13. The antibody or binding fragment thereof of claim 10 , wherein the antibody is a monoclonal or polyclonal antibody and wherein the antibody is further optionally chimeric or humanized.

14. The antibody or binding fragment of claim 10 , wherein the antibody fragment is a Fab, Fab′, F(ab′)2, scFv, dsFv, ds-scFv, dimer, minibody, diabody, or multimer thereof or a bispecific antibody fragment.

15. The antibody or binding fragment of claim 10 , wherein the antibody or binding fragment thereof is a monoclonal antibody or a binding fragment thereof.

16. A method for preparing a conformation specific antibody according to claim 1 , the method comprising immunizing a non-human subject with an immunogen or a composition comprising said immunogen and isolating an antibody that specifically binds EHAEVVFTA (SEQ ID NO:1).

17. The method of claim 16 further comprising forming an antibody-producing hybridoma.

A method of detecting a misfolded TTR conformation in a test sample, the method comprising contacting the test sample with a conformation specific antibody or binding fragment thereof of claim 1 under conditions suitable to form a specific antibody antigen complex between the antibody and TTR and determining if a specific antibody TTR complex is formed, thereby detecting misfolded TTR in the test sample.

The method of claim 18 , wherein the determining comprises measuring the amount of the antibody-antigen complex in the test sample and comparing the amount of antibody-antigen complex in the test sample to a control, wherein a difference in the amount of antibody-antigen complex in the test sample as compared to the control is indicative that misfolded TTR is detected in the test sample.

20. The method of claim 18 , wherein the test sample is a sample obtained from a subject having a TTR amyloid deposition disease.

21. The method of claim 20 , wherein the TTR amyloid deposition disease is selected from the group consisting of senile systemic amyloidosis, familial amyloid polyneuropathy and familial amyloid cardiomyopathy.

22. The method of claim 20 , wherein the test sample is a biological sample obtained from a subject, wherein the biological sample comprises blood, serum, plasma and/or solid tissue.

A method of inhibiting TTR fibril formation, the method comprising contacting a conformation specific antibody or fragment of claim 1 with misfolded TTR or monomeric TTR in solution under fibril formation conditions, wherein the antibody selectively binds the misfolded fibrils and inhibits fibril formation.
