Abstract

Described herein are methods of preventing, arresting progression of or ameliorating vision loss and other conditions associated with retinitis pigmentosa and x-linked retinitis pigmentosa in a subject. The methods include administering to a subject an effective concentration of a composition comprising a recombinant adeno-associated virus (AAV) carrying a nucleic acid sequence encoding a normal retinitis pigmentosa GTPase regulator (RPGR gene), or fragment thereof, under the control of regulatory sequences which express the product of the gene in the photoreceptor cells of the subject, and a pharmaceutically acceptable carrier.

Claims (12)

1. A recombinant adeno-associated virus (AAV) carrying a nucleic acid sequence encoding a human retinitis pigmentosa GTPase regulator (RPGR) gene product, under the control of regulatory sequences which express the gene product in the photoreceptor cells of a subject in need thereof, wherein the nucleic acid sequence comprises the sequence of SEQ ID NO: 1.

2. The AAV according to claim 1 , wherein the regulatory sequences comprise the interphotoreceptor retinoid-binding protein proximal promoter or G-protein-coupled receptor protein kinase 1 promoter.

3. The AAV according to claim 1 , wherein the AAV is an AAV2/5 pseudotyped AAV.

A recombinant adeno-associated virus (AAV) carrying a nucleic acid sequence encoding a human retinitis pigmentosa GTPase regulator (RPGR) gene product, under the control of regulatory sequences which express the gene product in the photoreceptor cells of a subject in need thereof, wherein the nucleic acid sequence encodes the sequence of SEQ ID NO: 5.

5. The AAV according to claim 1 , wherein the AAV is an AAV2.

A recombinant adeno-associated virus (AAV) carrying a nucleic acid sequence encoding a human retinitis pigmentosa GTPase regulator (RPGR) gene product, under the control of regulatory sequences which express the gene product in the photoreceptor cells of a subject in need thereof, wherein the nucleic acid sequence comprises a codon optimized variant of the sequence of SEQ ID NO:1, which encodes the sequence of SEQ ID NO: 5.

7. The AAV according to claim 6 , wherein the regulatory sequences comprise the interphotoreceptor retinoid-binding protein proximal promoter or G-protein-coupled receptor protein kinase 1 promoter.

8. The AAV according to claim 6 , wherein the AAV is an AAV2/5 pseudotyped AAV.

9. The AAV according to claim 6 , wherein the AAV is an AAV2.

10. The AAV according to claim 4 , wherein the regulatory sequences comprise the interphotoreceptor retinoid-binding protein proximal promoter or G-protein-coupled receptor protein kinase 1 promoter.

11. The AAV according to claim 4 , wherein the AAV is an AAV2/5 pseudotyped AAV.

12. The AAV according to claim 4 , wherein the AAV is an AAV2.
