Abstract

The invention provides proteins from group B streptococcus ( Streptococcus agalactiae ) and group A streptococcus ( Streptococcus pyogenes ), including amino acid sequences and the corresponding nucleotide sequences. Data are given to show that the proteins are useful antigens for vaccines, immunogenic compositions, and/or diagnostics. The proteins are also targets for antibiotics.

Claims (28)

1. An isolated nucleic acid molecule comprising a sequence selected from the group consisting of: (a) a sequence encoding a polypeptide comprising SEQ ID NO: 4212, and having at least one nucleotide modification to SEQ ID NO: 4211; and (b) a sequence encoding a fusion polypeptide, said fusion polypeptide comprising (i) a polypeptide which has at least 95% sequence identity to SEQ ID NO: 4212 and (ii) a heterologous polypeptide sequence.

2. The isolated nucleic acid molecule of claim 1 , comprising the nucleic acid sequence encoding the fusion polypeptide, said fusion polypeptide comprising (i) the polypeptide which has at least 95% sequence identity to SEQ ID NO: 4212 and (ii) the heterologous polypeptide sequence.

3. The isolated nucleic acid molecule of claim 1 , comprising the nucleic acid sequence encoding the fusion polypeptide, said fusion polypeptide comprising (i) the polypeptide which has at least 99% sequence identity to SEQ ID NO: 4212 and (ii) the heterologous polypeptide sequence.

4. The isolated nucleic acid molecule of claim 1 , comprising the nucleic acid sequence encoding the fusion polypeptide, said fusion polypeptide comprising (i) SEQ ID NO: 4212 and (ii) the heterologous polypeptide sequence.

5. An isolated nucleic acid molecule comprising a sequence that is fully complementary to the sequence of claim 1 .

6. An isolated a vector comprising a heterologous nucleic acid sequence and:

a) the nucleic acid sequence of claim 1 ;

b) the nucleic acid sequence of SEQ ID NO: 4211;

c) a nucleic acid sequence encoding a polypeptide comprising SEQ ID NO: 4212; or

d) a nucleic acid sequence encoding the polypeptide comprising SEQ ID NO: 4212, and having 95% or greater sequence identity to SEQ ID NO: 4211.

7. The isolated vector of claim 6 , wherein the heterologous nucleic acid sequence comprises a promoter operably linked to said nucleic acid sequence.

8. An isolated host cell comprising the isolated nucleic acid molecule of claim 1 .

9. The isolated host cell of claim 8 , wherein the cell is a mammalian cell, a bacterial cell other than a Streptococcus pyogenes cell or a Streptococcus agalactiae cell, or an Escherichia coli cell.

A composition comprising: (a) the isolated nucleic acid molecule of claim 1 , an isolated vector comprising said nucleic acid sequence, an isolated host cell comprising said isolated nucleic acid molecule or said vector, a polypeptide encoded by said nucleic acid sequence, or an isolated cell comprising said polypeptide, and (b) a pharmaceutically acceptable vehicle.

11. The composition of claim 10 , further comprising a polycationic agent and/or wherein the component of (a) is encapsulated in a lipid or packaged in a liposome.

12. An isolated host cell comprising the isolated vector of claim 6 .

13. The isolated host cell of claim 12 , wherein the cell is a mammalian cell, a bacterial cell other than a Streptococcus pyogenes cell or a Streptococcus agalactiae cell, or an Escherichia coli cell.

14. A composition comprising: (a) the isolated vector of claim 6 or an isolated host cell comprising said vector, and (b) a pharmaceutically acceptable vehicle.

15. The composition of claim 14 , further comprising a polycationic agent and/or wherein the component of (a) is encapsulated in a lipid or packaged in a liposome.

16. The isolated nucleic acid molecule of claim 1 , comprising the sequence encoding the polypeptide comprising SEQ ID NO: 4212, and having at least one nucleotide modification to SEQ ID NO: 4211.

17. The isolated nucleic acid molecule of claim 16 , comprising the sequence encoding the polypeptide comprising SEQ ID NO: 4212, and having at least 99% sequence identity to the sequence of SEQ ID NO: 4211.

18. The isolated vector of claim 6 , comprising the nucleic acid sequence of claim 1 .

19. The isolated vector of claim 6 , comprising the nucleic acid sequence of SEQ ID NO: 4211.

20. The isolated vector of claim 6 , comprising the nucleic acid sequence encoding the polypeptide comprising SEQ ID NO: 4212.

21. The isolated vector of claim 6 , comprising the nucleic acid sequence encoding the polypeptide of SEQ ID NO: 4212, and having 95% or greater sequence identity to SEQ ID NO: 4211.

22. The isolated vector of claim 21 , wherein the heterologous nucleic acid sequence comprises a promoter operably linked to said nucleic acid sequence.

23. An isolated host cell comprising the isolated vector of claim 21 .

24. An isolated host cell comprising the isolated vector of claim 20 .
