Abstract

Methods and compositions are provided for engineering mutant enzymes with reduced star activity where the mutant enzymes have a fidelity index (FI) in a specified buffer that is greater than the FI of the non-mutated enzyme in the same buffer.

Claims (16)

1. A composition comprising a variant SpeI restriction endonuclease having reduced star activity, wherein the variant SpeI restriction endonuclease comprises an amino acid sequence that differs from the amino acid sequence of the parent SpeI restriction endonuclease by an amino acid substitution at the position corresponding to position 65, in SEQ ID NO:17.

2. The composition according to claim 1 , wherein the amino acid substitution is P65A.

3. A composition comprising a variant SpeI restriction endonuclease having reduced star activity, wherein the variant SpeI restriction endonuclease comprises an amino acid sequence that differs from the amino acid sequence of the parent SpeI restriction endonuclease by a substitution at the amino acid position corresponding to position 59 in SEQ ID NO:17.

4. The composition according to claim 3 , wherein the substitution is E59A.

5. A composition comprising a variant SpeI restriction endonuclease having reduced star activity, wherein the variant SpeI restriction endonuclease comprises an amino acid sequence that differs from the amino acid sequence of the parent SpeI restriction endonuclease by a substitution at the amino acid position corresponding to position 108 in SEQ ID NO:17.

6. The composition according to claim 5 , wherein the substitution is S108A.

A composition comprising a variant SpeI restriction endonuclease having reduced star activity, wherein the variant SpeI restriction endonuclease comprises an amino acid sequence that differs from the amino acid sequence of the parent SpeI restriction endonuclease by a substitution at the amino acid position corresponding to position 172 in SEQ ID NO:17.

8. The composition according to claim 7 , wherein the substitution is N172A.

9. A composition comprising a variant SpeI restriction endonuclease having reduced star activity, wherein the variant SpeI restriction endonuclease comprises an amino acid sequence that differs from the amino acid sequence of the parent SpeI restriction endonuclease by a substitution at the amino acid position corresponding to position 174 in SEQ ID NO:17.

10. The composition according to claim 9 , wherein the substitution is K174A.

11. A composition comprising a variant SpeI restriction endonuclease having reduced star activity, wherein the variant SpeI restriction endonuclease comprises an amino acid sequence that differs from the amino acid sequence of the parent SpeI restriction endonuclease by a substitution at the amino acid position corresponding to position 179 in SEQ ID NO:17.

12. The composition according to claim 11 , wherein the substitution is Q179A.

13. A composition comprising a variant SpeI restriction endonuclease having reduced star activity, wherein the variant SpeI restriction endonuclease comprises an amino acid sequence that differs from the amino acid sequence of the parent SpeI restriction endonuclease by a substitution at the amino acid position corresponding to position 182 in SEQ ID NO:17.

14. The composition according to claim 13 , wherein the substitution is G182A.

15. A composition comprising a variant SpeI restriction endonuclease having reduced star activity, wherein the variant SpeI restriction endonuclease comprises an amino acid sequence that differs from the amino acid sequence of the parent SpeI restriction endonuclease by a substitution at the amino acid position corresponding to position 55 in SEQ ID NO:17.

16. The composition according to claim 15 , wherein the substitution is Y55F.
