Abstract

Described herein are compounds that are estrogen receptor modulators. Also described are pharmaceutical compositions and medicaments that include the compounds described herein, as well as methods of using such estrogen receptor modulators, alone and in combination with other compounds, for treating diseases or conditions that are mediated or dependent upon estrogen receptors.

Claims (38)

1. A compound of Formula (VII) , or a pharmaceutically acceptable salt thereof:

wherein:

X is —O—;

ring A is monocyclic heteroaryl or bicyclic heteroaryl;

ring B is phenyl or a monocyclic heteroaryl;

ring T is a fused phenyl, a fused monocyclic heteroaryl, or a fused bicyclic heteroaryl;

R 1 is H, F, C 1 -C 4 alkyl, or C 1 -C 4 fluoroalkyl;

each R 4 is independently, halogen, —CN, —OH, —OR 9 , —SR 9 , —S(═O)R 10 , —S(═O) 2 R 10 , —NHS(═O) 2 R 10 , substituted or unsubstituted C 1 -C 6 alkyl, substituted or unsubstituted C 1 -C 6 fluoroalkyl, substituted or unsubstituted C 1 -C 6 fluoroalkoxy, substituted or unsubstituted C 1 -C 6 alkoxy, or substituted or unsubstituted C 1 -C 6 heteroalkyl;

each R 5 is independently H, halogen, —CN, —OH, —OR 9 , —SR 9 , —S(═O)R 10 , —S(═O) 2 R 10 , —NHS(═O) 2 R 10 , substituted or unsubstituted C 1 -C 6 alkyl, substituted or unsubstituted C 1 -C 6 fluoroalkyl, substituted or unsubstituted C 1 -C 6 fluoroalkoxy, substituted or unsubstituted C 1 -C 6 alkoxy, or substituted or unsubstituted C 1 -C 6 heteroalkyl;

each R 6 is independently H, halogen, —CN, —OH, —OR 9 , —SR 9 , —S(═O) 2 R 10 , —S(═O) 2 R 10 , —NHS(═O) 2 R 10 , substituted or unsubstituted C 1 -C 6 alkyl, substituted or unsubstituted C 1 -C 6 fluoroalkyl, substituted or unsubstituted C 1 -C 6 fluoroalkoxy, substituted or unsubstituted C 1 -C 6 alkoxy, or substituted or unsubstituted C 1 -C 6 heteroalkyl;

R 7 is H, halogen, CN, substituted or unsubstituted C 1 -C 6 alkyl, or substituted or unsubstituted C 1 -C 6 fluoroalkyl;

each R 9 is independently H, —C(═O)R 10 , —C(═O)OR 10 , —C(═O)NHR 10 , substituted or unsubstituted C 1 -C 6 alkyl, substituted or unsubstituted C 1 -C 6 heteroalkyl, substituted or unsubstituted C 1 -C 6 fluoroalkyl, substituted or unsubstituted C 3 -C 10 cycloalkyl, substituted or unsubstituted C 2 -C 10 heterocycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted heteroaryl, —C 1 -C 2 alkylene-(substituted or unsubstituted C 3 -C 10 cycloalkyl), -C 1 -C 2 alkylene-(substituted or unsubstituted C 2 -C 10 heterocycloalkyl), —C 1 -C 2 alkylene-(substituted or unsubstituted aryl), or —C 1 -C 2 alkylene-(substituted or unsubstituted heteroaryl); or

each R 10 is independently substituted or unsubstituted C 1 -C 6 alkyl, substituted or unsubstituted C 1 -C 6 heteroalkyl, substituted or unsubstituted C 1 -C 6 fluoroalkyl, substituted or unsubstituted C 3 -C 10 cycloalkyl, substituted or unsubstituted C 2 -C 10 heterocycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted heteroaryl, —C 1 -C 2 alkylene-(substituted or unsubstituted C 3 -C 10 cycloalkyl), —C 1 -C 2 alkylene-(substituted or unsubstituted C 2 -C 10 heterocycloalkyl), —C 1 -C 2 alkylene-(substituted or unsubstituted aryl), or —C 1 -C 2 alkylene -(substituted or unsubstituted heteroaryl);

R 12 is H;

m is 0, 1, 2, 3, 4 or 5;

n is 0, 1, 2, 3 or 4;

p is 0, 1, 2, 3 or 4;

each R 8 is independently H, F, Cl, —OH, —CH 3 , —CH 2 CH 3 , —CH(CH 3 ) 2 , —CH 2 F, —CHF 2 , —CF 3 , —CH 2 CH 2 F, —CH 2 CHF 2 , —CH 2 CF 3 , —OCF 3 , —OCH 2 CF 3 , —OCH 3 , —OCH 2 CH 3 , —CH 2 OCH 3 , —CH 2 OCH 2 CH 3 , or —CH 2 OH.

2. The compound of claim 1 , or a pharmaceutically acceptable salt-thereof, wherein the compound has the structure of Formula (VIII):

3. The compound of claim 1 , or a pharmaceutically acceptable salt-thereof, wherein:

ring A is pyrrolyl, imidazolyl, pyrazolyl, triazolyl, oxazolyl, thiazolyl, thiophenyl, furanyl, pyridinyl, pyridazinyl, pyrazinyl, pyrimidinyl, phenyl, indazolyl, benzimidazolyl, benzotriazolyl, or benzoxazolyl;

each R 4 is independently H, halogen, —CN, —OH, —OR 9 , —SR 9 , —S(═O) 2 R l0 , -S(═O) 2 R 10 , C 1 -C 4 alkyl, C 1 -C 4 fluoroalkyl, C 1 -C 4 fluoroalkoxy, C 1 -C 4 alkoxy, or C 1 -C 4 heteroalkyl;

each R 5 is independently H, halogen, —OH, —OR 9 , —SR 9 , —S(═O)R 10 , —S(═O) 2 R 10 , C 1 -C 4 alkyl, C 1 -C 4 fluoroalkyl, C 1 -C 4 fluoroalkoxy, C 1 -C 4 alkoxy, or C 1 -C 4 heteroalkyl;

each R 6 is independently H, halogen, —OH, C 1 -C 4 alkyl, C 1 -C 4 fluoroalkyl, C 1 -C 4 fluoroalkoxy, or C 1 -C 4 alkoxy;

R 7 is C 1 -C 4 alkyl or C 1 -C 4 fluoroalkyl; and

p is 0, 1, or 2.

4. The compound of claim 1 , or a pharmaceutically acceptable salt-thereof, wherein:

or a pharmaceutically acceptable salt thereof.

6. A pharmaceutical composition comprising a therapeutically effective amount of a compound of claim 1 or 5 , or a pharmaceutically acceptable salt thereof.

7. The pharmaceutical composition of claim 6 , wherein the pharmaceutical composition is formulated for intravenous injection, subcutaneous injection, oral administration, or topical administration.

8. The pharmaceutical composition of claim 6 , wherein the pharmaceutical composition is a tablet, a pill, a capsule, a liquid, a suspension, a gel, a dispersion, a solution, an emulsion, an ointment, or a lotion.

9. The pharmaceutical composition of claim 6 wherein the pharmaceutical composition further comprises one or more additional therapeutically active agents.

10. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein:

R 1 is H or —CH 3 ; and R 7 is CH 3 .
